摘要
AMEP蛋白是从枯草芽孢杆菌中分离鉴定的一种新型蛋白激发子,能够激发植物免疫,提高植物抗病性。经生物信息学分析发现,AMEP蛋白的编码基因在芽孢杆菌属的菌株中广泛分布,但各菌株AMEP蛋白的表达产量和活性还有待研究。该研究对实验室保藏的多株芽孢杆菌进行PCR扩增鉴定基因,筛选出含有AMEP编码基因的菌株;通过蛋白表达纯化和定量确定了各菌株AMEP蛋白的表达量;并通过过敏反应试验比较各菌株AMEP蛋白的功能活性,确定菌株BU396产AMEP蛋白的表达量最高,且具有正常的功能活性,适合于作为今后AMEP蛋白的表达菌株。
AMEP protein is a new protein elicitor isolated and identified from Bacillus subtilis,which can stimulate plant immunity and promote plant disease resistance.Bioinformatics analysis showed that the coding gene of AMEP protein was widely distributed in Bacillus strains,but the expression level and activity of AMEP protein in individual strain remained to be studied.In this paper,the strains containing AMEP coding gene were identified by PCR ampli⁃fication.Then the expression level of AMEP protein in each strain was determined by protein expression and purifica⁃tion.Finally,the activity of AMEP protein produced by each strain was determined by hypersensitive reaction assay.BU396 was screened out for both good expression level and high activity,which was suitable for the future applica⁃tion.
