摘要
目的探讨苦参碱对非小细胞肺癌(NSCLC)放射增敏作用机制。方法人NSCLC细胞株H1299、A549分为对照组、苦参碱组、放疗组、苦参碱+放疗组,分别用CCK-8、流式细胞术、平板克隆形成、蛋白印迹检测细胞增殖、凋亡、放射增敏、γ-H2AX表达。结果 0.25~8 g/L苦参碱处理明显抑制NSCLC细胞存活(P<0.01)。苦参碱+放疗组中细胞凋亡、γ-H2AX蛋白表达明显高于苦参碱组、放疗组,而细胞克隆数明显降低(P<0.01或0.05);H1299和A549细胞的放射增敏比分别为1.22和2.02。结论苦参碱联合放疗可能通过增加NSCLC细胞DNA损伤、诱导细胞凋亡,进而发挥放射增敏作用。
Objective To investigate the radiosensitizing mechanism of matrine on non-small cell lung cancer(NSCLC).Methods Human NSCLC cell lines H1299 and A549 were divided into control,matrine,radiotherapy,and matrine+radiotherapy groups,and their proliferation,apoptosis,radiosensitization,andγ-H2AX expression were respectively determined by CCK-8,flow cytometry,colony formation,and Western blot.Results Matrine at doses of 0.25-8 g/L significantly inhibited NSCLC cell viability(P<0.01).Compared with matrine and radiotherapy groups,apoptosis andγ-H2AX expression were increased but cell clones decreased(P<0.01 or 0.05)in matrine+radiotherapy group.The sensitization enhancement ratios of H1299 and A549 cells were respectively 1.22 and 2.02 after treated with matrine+radiotherapy.Conclusion Matrine may enhance radiosensitivity by induction of DNA damage and apoptosis in NSCLC cells.
