摘要
目的研究老年轻度认知功能障碍(Mild cognitive impairment,MCI)患者血清脑源性神经营养因子(Brain derived neurotrophic factor,BDNF)水平的变化,探讨两者间的相关性。方法从课题组既往完成的流行病学调查人群中选出汉族MCI患者100例,正常对照汉族老年人145例,使用上海Lengton生物公司BDNF检测试剂盒,采用酶联免疫吸附法(Enzyme linked immunosorbent assay,ELISA)测定MCI和正常老人血清BDNF水平进行统计学分析。结果 MCI组和正常对照组血清BDNF水平分别为3.14±1.71ng/ml和3.72±1.87ng/ml,MCI组血清BDNF水平低于正常对照组,差异有统计学意义(t=2.32,P=0.023);进一步线性相关分析显示MCI组血清BDNF水平与Moca量表总评分呈正相关关系(r=0.141,P=0.028);各维度分析显示血清BDNF浓度与Moca量表中注意力和计算力、回忆能力、定向力等维度均呈正相关关系(P=0.048,0.009,0.031);与视空间与执行能力、命名能力、语言能力等维度未发现有相关性。结论 MCI患者的血清BDNF水平较正常对照降低,BDNF与MCI患者计算力、回忆能力和定向力有关,提示BDNF水平的下降可能参与了MCI患者病理损害的过程,影响了MCI的发生和发展。
Objective To investigate the relationship between brain-derived neurotrophic factor(BDNF) and mild cognitive impairment in elder patients. Methods 100 cases with mild cognitive impairment(MCI) and 145 control normal elders were selected from the MCI epidemiological survey in Xin jiang. BDNF dectection kit made by Shanghai Lengton biotechnology company were used in this study. The cognitive function was evaluated by neuropsychological test and the level of BDNF was tested by enzyme linked immunosorbent assay ELISA. Results The serum levels of BDNF in MCI group and control group were 3.14±1.71 ng/ml and 3.72±1.87 ng/ml respectively. Compared with the control group, the level of BDNF in MCI patients was significantly decreased(t = 2.32,P =0.023). Further linear correlation analysis showed that serum BDNF level in MCI group was positively correlated with total Moca scale score(r = 0.141,P = 0.028). The analysis of all dimensions showed that serum BDNF concentration was positively correlated with attention, computational power, recall ability and orientation in the Moca scale(P = 0.048,0.009,0.031). No correlation was found between visual space and executive ability, naming ability, language ability and other dimensions. Conclusions The serum BDNF level of MCI patients was lower than that of the normal control, and BDNF was related to the computational power, recall ability and orientation ability of MCI patients. The decreased level of BDNF in MCIpatients showed that BDNF may be involved in the pathophysiology of cognitive impairment and has effect on the occurence and development of MCI.
