摘要 [目的]探讨热休克蛋白90对猪胚胎发育过程的影响.[方法]采用改良的退火控制引物差分显示(ACP)技术检测人工授精和核移植猪胚胎中的差异表达基因,再利用实时定量PCR法确认基因的表达情况.[结果]通过改良的ACP技术获得了若干个差异表达基因,利用实时定量PCR法进行了进一步确认,结果见热休克蛋白90在核移植猪胚胎中的表达水平明显低于人工授精猪胚胎(P<0.01).[结论]热休克蛋白90可能参与猪胚胎的发育过程. OBJECTIVE To explore the effects of heat shock protein 90(HSP90) on the development of porcine embryo. METHODS The differentially expressed genes(DEGs) in porcine in vitro fertilization(IVF) and nuclear transfer(NT) blastocysts were detected by improved annealing control primers(ACPs) differential display technique, and the expressions of DEGs were confirmed by real-time quantitative PCR. RESULTS Several DEGs were obtained by improved ACP technique and further confirmed by real-time quantitative PCR, the results showed that the expression of HSP90 in NT porcine embryos was significantly lower than that in IVF porcine embryos(P<0.01). CONCLUSION HSP90 may be involved in the development of porcine embryos.
