HPLC同时测定补益地黄丸中的7种成分 被引量:4

Simultaneous determination of seven compositions in Buyi Dihuang pills by HPLC

摘要 目的采用HPLC法同时测定补益地黄丸中毛蕊花糖苷、焦地黄苯乙醇苷B1、β-蜕皮甾酮、芸香柚皮苷、柚皮苷、橙皮苷和新橙皮苷的含量。方法采用Agilent Extend XDB-C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%磷酸溶液,梯度洗脱,流速1.0 mL·min^-1,柱温30℃,检测波长分别为330 nm(0~24.0 min,毛蕊花糖苷、焦地黄苯乙醇苷B1)、250 nm(24.0~33.0 min,β-蜕皮甾酮)、283 nm(33.0~70.0 min,芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷)。结果毛蕊花糖苷、焦地黄苯乙醇苷B1、β-蜕皮甾酮、芸香柚皮苷、柚皮苷、橙皮苷和新橙皮苷的线性范围分别为2.67~53.40、1.09~21.80、0.86~17.20、2.97~59.40、16.59~331.80、1.48~29.60、7.56~151.20μg·mL^-1(r≥0.9991),平均加样回收率分别为98.90%、97.76%、96.97%、99.35%、100.02%、98.10%、99.85%,RSD分别为1.11%、1.38%、0.98%、0.79%、1.05%、1.28%、0.59%。结论所用方法简便、重复性好,可用于补益地黄丸的质量控制。 OBJECTIVE To establish an HPLC method for the simultaneous detemination of verbascoside,jionoside B1,β-ecdysterone,narirutin,naringin,hesperidin and neohesperidin in Buyi Dihuang pills.METHODS The detemination was performed on Agilent Extend XDB-C18(250 mm×4.6 mm,5μm)column with a mobile phase consisting of acetonitrile-0.2%phosphoric acid solution by a gradient elution.The flow rate was 1.0 mL·min^-1 and the column temperature was 30℃.The detection wavelengths were set at 330 nm for verbascoside and jionoside B1 at 0-24.0 min;250 nm forβ-ecdysterone at 24.0-33.0 min;283 nm for narirutin,naringin,hesperidin and neohesperidin at 33.0-70.0 min.RESULTS Verbascoside,jionoside B1,β-ecdysterone,narirutin,naringin,hesperidin and neohesperidin showed good linear relationships within the ranges of 2.67-53.40,1.09-21.80,0.86-17.20,2.97-59.40,16.59-331.80,1.48-29.60 and 7.56-151.20μg·mL^-1(r≥0.9991),respectively.The average recoveries were 98.90%,97.76%,96.97%,99.35%,100.02%,98.10%and 99.85%with the RSDs of 1.11%,1.38%,0.98%,0.79%,1.05%,1.28%and 0.59%,respectively.CONCLUSION The method is easy and repeatable,which can be used for quality control of Buyi Dihuang pill.
