不同生长年限大血藤药材中7种成分的含量比较 被引量:5

Comparison of seven components in Sargentodoxae Caulis with different growing years

摘要 目的研究不同生长年限大血藤药材中7种活性成分的动态变化,为大血藤的标准化种植、采收及质量控制提供参考。方法采用高效液相色谱法,分别测定不同生长年限的人工栽培及野生大血藤中7种成分的含量;采用Agilent Extend C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.2%乙酸水溶液,梯度洗脱,检测波长280 nm(红景天苷、没食子酸和原儿茶酸)、254 nm(大黄素)、327 nm(咖啡酸、香草酸和绿原酸),流速1.0 mL·min^-1,柱温30℃。结果1~5年生人工栽培大血藤中没食子酸、原儿茶酸、绿原酸、咖啡酸、香草酸、红景天苷、大黄素的含量分别为0.3803~1.3625、0.2558~1.7718、0.6023~6.7012、0.0842~2.7145、0.2661~2.3415、0.0447~0.9014、0.0175~0.4327 mg·g^-1。7种成分在测定浓度范围内的线性关系良好,r均大于0.9994,精密度、重复性和稳定性良好;平均回收率为98.43%~101.90%,RSD为0.89%~1.75%。结论不同生长年限的大血藤中7种成分的含量差异明显,1~3年生大血藤中7种成分的含量较低,4年生的含量显著升高,4~5年生的含量趋于稳定;说明大血藤种植4年即可采收。 OBJECTIVE To study dynamic changes of seven active components of Sargentodoxae Caulis with different growth years,and to provide references for the standardized planting,harvesting and quality control.METHODS The content of seven kinds in wild and artificial cultivation of Sargentodoxae Caulis was determined by HPLC.The column was Agilent Extend C18(250 mm×4.6 mm,5μm).The mobile phase was methanol-0.2%phosphoric acid with gradient elution.The detection wavelength was 280 nm for salidroside,callic acid and protocatechuic acid,254 nm for emodin,and 327 nm for caffeic acid,vanillic acid and chlorogenic acid.The flow rate was 1.0 mL·min^-1 with the column temperature of 30℃.RESULTS The contents of callic acid,protocatechuic acid,chlorogenic acid,caffeic acid,vanillic acid,salidroside and emodin in artificial cultivation of Sargentodoxae Caulis from 1-5 years were 0.3803-1.3625 mg·g^-1,0.2558-1.7718 mg·g^-1,0.6023-6.7012 mg·g^-1,0.0842-2.7145 mg·g^-1,0.2661-2.3415 mg·g^-1,0.0447-0.9014 mg·g^-1 and 0.0175-0.4327 mg·g^-1,respectively.The linear relation of seven components in the range of mass concentration was good with perfect precision,repeatability and stability,the value of which was all more than 0.9994.The average recovery rate was 98.43%-101.90%with the RSD of 0.89%-1.75%.CONCLUSION The diversity of the contents from different growing years is great.The seven contents in Sargentodoxae Caulis in 1-3 years are in the lower levels,and the ingredients in 4 years increased significantly and the ingredients in 4-5 years have stable change.Sargentodoxae Caulis should be harvested after 4 years’growth.
