寻常性银屑病表皮HLA-G甲基化水平的检测 被引量:2

摘要 目的通过检测寻常性银屑病患者表皮人类白细胞抗原-G(human leukocyte antigen-G,HLA-G)基因启动子区甲基化水平,研究HLA-G基因启动子区甲基化对银屑病发生发展的影响。方法采用亚硫酸氢钠测序技术检测HLA-G基因启动子区甲基化水平,将银屑病患者皮损部位、非皮损部位及健康正常人表皮进行对照,分析其与病情严重程度的相关性。以银屑病皮损面积及严重程度指数(PASI)评估银屑病病情严重程度。结果在56例银屑病患者皮损、非皮损部位及28名健康对照者表皮中检测到不同程度的HLA-G基因启动子区甲基化,分别为(36.69±7.92)%、(37.38±6.90)%和(57.23±11.89)%,银屑病患者皮损及非皮损部位HLA-G基因启动子区甲基化水平均显著低于健康正常人(P<0.001),皮损及非皮损部位HLA-G启动子区甲基化水平比较差异无统计学意义(P>0.05)。皮损部位HLA-G启动子区甲基化水平与银屑病的严重程度不相关(r=-0.24,P>0.05)。结论银屑病患者表皮中HLA-G基因启动子区甲基化水平明显降低,可能是银屑病患者的一种表观遗传标志。 Objective To investigate the role of promoter methylation of human leukocyte antigen(HLA)-G in the development of psoriasis by detecting the promoter methylation levels of HLA-G in the psoriatic epidermis in patients with psoriasis vulgaris.Methods Sodium Bisulfite Sequencing was applied to detect the promoter methylation levels of HLA-G in psoriatic lesions and the non-lesional areas from 56 patients with psoriasis vulgaris,and normal epidermis from 28 healthy controls.We compared and evaluated the correlation between the promoter methylation levels of HLA-G and the severity of the disease.The severity of psoriasis was assessed by PASI.Results Different degrees of promoter methylation levels of HLA-G were detected in the lesional and non-lesional epidermis of 56 patients with psoriasis vulgaris and 28 healthy controls,at(36.69±7.92)%,(37.38±6.90)%,(57.23±11.89)%,respectively.The promoter methylation levels of HLA-G in both lesional and non-lesional epidermis from the patients with psoriasis were significantly lower than healthy controls(P<0.001).There is no significant difference for HLA-G methylation levels between lesional and non-lesional epidermis from the patients with psoriasis(P>0.05).However,the methylation levels of HLA-G lesional and nonlesional epidermis from the patients with psoriasis are not related to the severity of psoriasis(P>0.05).Conclusion The promoter methylation levels of HLA-G in psoriatic epidermal lesions were significantly decreased,suggesting that promoter methylation levels of HLA-G could be an epigenetic marker in psoriasis.
