摘要
目的:建立合欢花药材HPLC指纹图谱分析方法,结合模式识别与含量测定为其质量控制提供科学依据。方法:采用HPLC法,Waters CORTECS C18色谱柱(150 mm×4.6 mm,2.7μm),以乙腈-0.1%磷酸水为流动相,梯度洗脱,流速1.0 m L/min,检测波长270 nm。通过相似度分析、主成分分析、正交偏最小二乘法判别分析对16批合欢花指纹图谱进行评价,寻找合欢花的主要差异性化学成分,并测定其含量。结果:建立了合欢花药材的指纹图谱,确定12个共有峰,并指认了其中3个共有峰,16批药材相似度均大于0.90。根据聚类分析结果,可将16批药材聚为2类,通过主成分分析和正交偏最小二乘法判别分析确定槲皮苷为合欢花的主要差异性化学成分,方法学考察符合规定,RSD均小于3.0%。结论:该方法精密度高、重现性好,可为合欢花药材质量控制与评价提供参考。
Objective To establish the HPLC fingerprint analysis method of flower of Albizia julibrissin,and to provide scientific basis for its quality control by combining pattern recognition and content determination.Methods HPLC method was adopted,the determination was performed on Waters CORTECS C18(150 mm×4.6 mm,2.7μm)column with mobile phase consisted of acetonitrile-0.1%phosphate water.The flow rate was 1.0 m L/min,and the detection wavelength was 270 nm.The fingerprints of 16 batches of flower of Albizia julibrissin samples were analyzed by similarity analysis,principal component analysis and orthogonal partial least squares discriminant analysis,the main different chemical component of flower of Albizia julibrissin was searched and the content determination was conducted.Results The HPLC fingerprint of flower of Albizia julibrissin was established,and 12 common peaks were calibrated.3 common peaks were identified,the similarity of 16 batches of medicinal materials were all higher than 0.90.Through cluster analysis,16 batches of medicinal materials could be clutered into 2 groups,quercitrin was identified as the main different chemical component of flower of Albizia julibrissin by principal component analysis and orthogonal partial least squares discriminant analysis,the methodological study in accordance with the provisions,RSD was less than 3.0%.Conclusion The method has high precision and reproducibility,which can provide reference for the quality control and evaluation of flower of Albizia julibrissin.
