摘要
以如意皇后粗肋草带侧芽的根茎为试验材料,研究植物激素对如意皇后粗肋草离体培养的影响,并对灭菌时间、侧芽诱导、愈伤组织诱导、丛生芽诱导、丛生芽增殖以及生根培养条件进行选择。研究表明:外植体灭菌18 min效果最佳,侧芽诱导效果最佳培养基为:MS+2.0 MS+2.0 mg/L 6-BA+0.4 mg/L IBA,愈伤组织诱导效果最佳培养基为:MS+1.0 mg/L TDZ+2.0 mg/L 2,4-D,1/2MS+0.6 mg/L TDZ,对丛生芽诱导效果最佳,丛生芽增殖最佳培养基MS+4.0 mg/L KT+0.1 mg/L NAA和MS+6.0 mg/L 6-BA+0.1 mg/L NAA培养效果最佳,培养基1/2MS+0.3 mg/L NAA生根培养效果最佳。通过研究,初步得到适宜外植体生长的培养基配方,为如意皇后粗肋草试管苗的培养提供技术支撑。
The rhizomes with side buds of Ruyi queen were used as test materials to study the effects of plant hormones on the in vitro culture of ruyi queen,and the sterilization time,lateral bud induction,callus induction,cluster bud induction,Cluster bud proliferation and rooting culture conditions.Studies have shown that the best effect of sterilization of the explants for 18 minutes and the best medium for lateral bud induction are:MS+2.0 MS+2.0 mg/L 6-BA+0.4 mg/L IBA,the best callus induction effect The medium was:MS+1.0 mg/L TDZ+2.0 mg/L 2,4-D,1/2MS+0.6 mg/L TDZ had the best effect on induction of cluster buds,and the best medium for cluster bud proliferation was MS+4.0 mg/L KT+0.1 mg/L NAA and MS+6.0 mg/L 6-BA+0.1 mg/L NAA have the best culture effect,and the medium 1/2MS+0.3 mg/L NAA has the best rooting culture effect.Through research,a medium formula suitable for the growth of explants was initially obtained,which was intended to provide technical support for the cultivation of Ruyi queens test tube seedlings.
