梨离体再生体系建立

摘要 黄金梨(Pyrus pyrifolia cv.Whangkumbe)属蔷薇科(Rosaceae)梨属(Pyrus L.)植物,在福建种植面积较为广泛。建立梨稳定、高效的再生体系,是进行转基因育种的关键环节之一。本试验以黄金梨成熟胚为试材,进行再生体系建立研究,通过萌发、增殖与生根培养,筛选出最佳培养基,探讨不同基本培养种类、植物生长调节剂对再生体系建立的影响,以期为梨良种扩繁提供基础。结果表明:最佳萌发培养基为1/2MS+6-BA 0.4 mg·L^-1,种子的发芽率为93.02%。最佳增殖培养基为:MS+6-BA 1.0 mg·L^-1+IBA 0.2 mg·L^-1+GA30.2 mg·L^-1,增殖系数为4.72。诱导生根最佳培养基为:1/4 MS+NAA 1.0 mg·L^-1+IBA 1.0 mg·L^-1+蔗糖20 g·L^-1,生根率为65.79%。 Pear cv.‘Whangkumbe’,plant of family Rosaceae and genus Pyrus,is widely grown in Fujian.A stable and efficient regeneration system is one of the key links of transgenic breeding.In this study the mature embryo of cv.‘Whangkumbe’was used as materials to build in vitro regeneration system.Through germination,proliferation and rooting culture,the best medium was screened and effects of different types of basic medium and plant growth regulators impact on in vitro regeneration system were discussed in order to lay a foundation for the improvement of pear tree cultivars.The results showed that the best medium for germination was 1/2MS+6-BA 0.4 mg·L^-1,and,the germination percentage was 93.02%.The best proliferation medium was MS+6-BA 1.0 mg·L^-1+IBA 0.2 mg·L^-1+GA30.2 mg·L^-1 with the proliferation coefficient of 4.72;the best medium for rooting was 1/4 MS+NAA 1.0 mg·L^-1+IBA 1.0 mg·L^-1+sucrose 20 g·L^-1 with the rooting rate of 65.79%.
