摘要
目的基于神经网络模型评价淫羊藿女贞子配伍对自然衰老大鼠肝脏及脑组织衰老的影响。方法3、6月龄雄性大鼠(各8只)作青年对照组;8月龄雄性SD大鼠56只,随机分为7组,每组8只,包括9月龄组、12月龄组、15月龄组、17月龄组、淫羊藿组、女贞子组、淫羊藿女贞子组(配伍组),饲养至相应月龄(各用药组饲养至15月龄后开始灌胃给予相应药物2个月)后处死。剖取肝及脑组织,HE染色观察肝脏病理改变;制备肝及大脑组织匀浆,检测总抗氧化能力(Total antioxidant capacity,T-AOC)、超氧化物歧化酶(Superoxide dismutase,SOD)、还原型谷胱甘肽(Glutathione,GSH)、一氧化氮(Nitric oxide,NO)及丙二醛(Malondial-dehyde,MDA)。结果与青年大鼠比较,17月龄组大鼠肝脏出现退行性的病理改变,肝及脑组织T-AOC、GSH及SOD水平显著降低,NO及MDA含量显著升高(P<0.05或P<0.01)。与17月龄组比较,淫羊藿组肝重、肝脏指数显著性降低,肝细胞面积显著升高(P<0.05或0.01);女贞子组肝重显著性降低,肝细胞面积显著升高(P<0.05);配伍组肝重及脂肪变性程度降低(P<0.05或P<0.01);各用药组肝脑中T-AOC、GSH、SOD、MDA和NO含量差异均有统计学意义(均P<0.01)。基于多层感知(MLP)神经网络模型各用药组月龄值,较17月龄组均有显著降低(均P<0.01),且合用组优于淫羊藿、女贞子单用组(P<0.05或P<0.01)。结论淫羊藿女贞子配伍延缓衰老的作用可能与提高衰老大鼠肝脑组织的抗氧化能力,减轻组织氧化损伤作用有关。
Objective To evaluate the effects of the combination of Herba Epimedii and Ligustrum lucidum on liver and brain of aging rats based on neural network model.Methods 3-month-old and 6-month-old male rats(8 rats in each group)were regarded as the youth control group.Fifty-six male SD rats aged 8 months were randomly divided into 7 groups,including 9-month-old group,12-monthold group,15-month-old group,17-month-old group(17 M group),Herba Epimedii group(H group),Ligustrum lucidum group(F group),and compatibility group(C group).After being fed to the correspondent age(treatment groups were fed to the age of 15 months normally following correspondent drugs administration by gavage for two months),the experimental rats were sacrificed and taken and the liver and brain.We observed pathological changes of liver by HE staining and grinded liver and brain tissue for homogenate to detect the following indicators of liver and brain:total antioxidant capacity(T-AOC),superoxide dismutase(SOD),glutathione(GSH),nitric oxide(NO),malondialdehyde(MDA).Results Compared with youth control group,degenerative pathological changes of liver can be observed,the levels of T-AOC,GSH and SOD were decreased significantly and the levels of NO and MDA were increased significantly(P<0.05 or P<0.01)in liver and brain tissue of rats in 17 M group.Compared with 17 M group,the weight and index of liver were significantly decreased and hepatocyte area were increased(P<0.05 or P<0.01)in H group;the weight of liver was decreased and hepatocyte area was increased(P<0.05)in F group;the weight and degree of steatosis of liver were decreased significantly(P<0.05 or P<0.01)in C group;the levels of T-AOC,GSH,SOD,MDA and NO in liver and brain tissue all had statistical differences(all P<0.01)in each treatment group.Basing on the multi-layer neural network(MLP)model,the results showed that there was a significant difference between of each treatment group and 17 M group in the predicted age(all P<0.01),and the effect of C group was better than H group and F group(P<0.05 or P<0.01).Conclusion The effects of the combination of Herba Epimedii and Ligustrum lucidum on delaying senescence might be related to improving the antioxidant capacity and reducing the oxidative damage of liver and brain tissue.
