甘薯渣多糖提取、分离纯化及抗氧化活性研究 被引量:11

摘要 研究甘薯渣多糖的提取,分离纯化的方法,并分析其体外抗氧化活性。结果表明:超声辅助水浴法提取甘薯渣多糖的最佳工艺为料水比1∶17、超声时间57 min、超声功率209 W、超声温度60℃,甘薯渣多糖的平均得率为5.926%。经Sevag法和木瓜蛋白酶联合脱蛋白、H2O2法脱色、透析、DEAE-52纤维素柱层析得到含量较多的甘薯渣精多糖SPRP-1。经纯度鉴定分析可知SPRP-1是一种不含蛋白质、核酸的较纯的多糖。对体外抗氧化活性分析得,SPRP-1对DPPH自由基、超氧阴离子自由基、·OH的清除能力较强。 In this paper,the extraction,separation and purification methods of polysaccharides from sweet potato residue were studied,and the antioxidant activity in vitro was analyzed.The results indicated that the optimal parameters for extracting polysaccharides from sweet potato residue by ultrasound-assisted water bath alcohol precipitation were 1∶17 in material-liquid ratio,ultrasonic time 57 minutes,ultrasonic power 209 W,ultrasonic temperature 60℃,and under these best conditions,average polysaccharide yield of sweet potato residue was 5.926%.Then Sevag method combined with papain was used to remove protein and H2O2 method was used to remove pigment.The main composition of sweet potato residue pure polysaccharides without proteins and nucleic acids were SPRP-1,which was purified by dialyzing and DEAE-52 cellulose column chromatography.Antioxidant activity by analyzing showed that SPRP-1 was efficient to scavenge DPPH free radicals,superoxide anion and hydroxy radicals.
