参苓白术散改善脂质代谢的生物信息分析及其与肠道菌群的关系 被引量:10

摘要 目的:基于网络药理生物信息技术分析,探讨参苓白术散治疗血脂异常的分子作用机制,同时,本研究亦探究参苓白术散促进脂质代谢可能与其重塑肠道微生物稳态相关,挖掘与菌群相关的可能分子机制。方法:采用中药系统药理学分析平台(TCMSP)对参苓白术散中化合物中活性成分和作用靶点进行筛选,通过检索Genecards数据库获取血脂异常疾病相关靶点,构建疾病-靶点-化合物网络;筛选药物靶点和疾病靶点相互作用的有效靶点;利用DAVID对有效作用靶点进行GO分析及KEGG Pathway富集分析。结果:根据OB≥30%及DL≥0.18,筛选出194个活性成分及127个可能蛋白靶点;通过Venn图获得63个参苓白术散治疗血脂异常的有效作用靶点;GO分析富集结果,根据P<0.05筛选前10个可能基因;利用KEGG数据库对相关通路富集,筛选前10条可能信号通路;筛选存在与肠道菌群参与调节脂质代谢相关的靶点,可能与其调控IL6、PPARA、MAPK1、APOB、TNF分子,及FoxO signaling pathway、MAPK signaling pathway、PPAR signaling pathway信号通路相关。结论:通过网络药理学的初步筛选及预测,参苓白术散可能通过多途径、多靶点改善脂质代谢,该方可能通过对肠道菌群的重塑进而促进脂质代谢。 Objective:To explore the molecular mechanism of Shenling Baishu Powder in the treatment of dyslipidemia based on network pharmacology and bioinformatics analysis.At the same time,this study also explored the possible relationship between the promotion of lipid metabolism and the remodeling of intestinal microbial homeostasis,and to explore the possible molecular mechanism related to the flora.Methods:Firstly,the active components and targets in Shening Baizhu Powder were screened and predicted by TCMSP.Then,targets related to dyslipidemia were obtained by searching Gene cards database,and the disease-gene-target-compound network was constructed.The effective drug target and disease target interaction target were screened.GO analysis and KEGG Pathway enrichment analysis were performed on the effective target by DAVID.Results:A total of 194 active components and 127 possible protein targets were screened according to OB≥30%and DL≥0.18.Through Venn figure,63 effective targets of Shenling Baizhu Powder in the treatment of dyslipidemia were obtained.Enrichment results of GO analysis were used to screen the first 10 possible genes according to P<0.05.KEGG database was used to enrich related pathways and screen the first 10 possible signaling pathways.The screening results showed that there were targets for regulating lipid metabolism with intestinal flora,which can be entered by IL6,PPARA,MAPK1,APOB,TNF,FoxO signaling pathway,MAPK signaling pathway,PPAR signaling pathway and other signaling pathways.Conclusion:Through the preliminary screening and prediction of network pharmacology,Shenling Baizhu Powder may improve lipid metabolism through multiple pathways and targets,further clarify the internal mechanism of the prescription to promote lipid metabolism by remodeling the intestinal flora.
