摘要
目的通过检测卵巢浆液性肿瘤(OST)组织中上皮型-钙粘蛋白(E-Cadherin)、β-连环蛋白(β-Catenin)、第10号染色体缺失的磷酸酶和张力蛋白同源物基因(PTEN)、磷脂酰肌醇-3激酶(PI3K)110α、磷酸化丝氨酸蛋白激酶B(p-PKB/AktSer473)以及Ki-67表达,揭示E-Cadherin/β-Catenin与PTEN-PI3K/Akt通路在OST发生发展中的作用,探讨OST的发生发展机制。方法应用免疫组化法,检测10例卵巢正常上皮(NOT)、12例浆液性囊腺瘤(OSA)、18例浆液性交界性囊腺瘤(OSBT)、60例卵巢浆液性癌(OSC)石蜡包埋组织中E-Cadherin、β-Catenin、PTEN、PI3K110α、p-AktSer 473以及Ki-67蛋白表达,并结合OSC的临床病理特征进行统计分析。结果(1)OSC组E-Cadherin、PTEN阴性细胞率均高于OSBT、OSA和NOT(均P<0.05)。(2)OSC浸润与非浸润的标本中E-Cadherin、PTEN阴性细胞率的差异有统计学意义(P<0.05);OSC有无淋巴结转移的标本中E-Cadherin、PTEN阴性细胞率差异也有统计学意义(P<0.05)。(3)OSC中PI3Kp110α、p-AktSer473、β-Catenin以及Ki-67阳性细胞率高于OSBT、OSA和NOT,差异有统计学意义(P<0.01)。(4)OSC浸润与非浸润的标本比较,PI3Kp110α、p-AktSer473、β-Catenin以及Ki-67阳性细胞率的差异有统计学意义(P<0.01);OSC有无淋巴结转移的标本比较,PI3Kp110α、p-AktSer473、β-Catenin以及Ki-67阳性细胞率的差异均有统计学意义(均P<0.01)。结论E-Cadherin、PTEN蛋白表达的缺失伴随PI3K110α、p-AktSer473、β-Catenin以及Ki-67蛋白的过度激活,可能共同参与OST的发生发展,检测上述蛋白可为OST的临床诊治以及预后判断提供依据。
Objective To investigate the expression of epithelial calcium-dependent cell adhesion proteins(E-Cadherin)/β-Catenin and phosphatase and tension homologdeleted on chromosome ten(PTEN)-phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt/PKB)in ovarian serous tumor(OST)and their clinicopathological significance.Methods The expressions of E-Cadherin,β-Catenin,PTEN,PI3K110α,p-AktSer473 and Ki-67 were detected by immunohistochemical method in 10 tissue samples of normal ovarian epithelia(NOT),12 samples of serous cystadenoma(OSA),18 samples of serous borderline cystadenoma(OSBT),and 60 samples of ovarian serous carcinoma(OSC).The relationship between expression of above proteins and the clinicopathological features was analyzed.Results The negative expression rates of E-Cadherin,PTEN in OSC group were significantly higher than those in OSBT,OSA and NOT groups(P<0.05).The negative rates of E-Cadherin and PTEN in infiltrated and non-infiltrating specimens of OSC group were statistically different(P<0.05).The negative rates of E-Cadherin and PTEN in the samples with or without lymph node metastasis of OSC group were also statistically different(P<0.05).The positive rates of PI3K110α,p-AktSer473,β-Catenin and Ki-67 protein in OSC group were significantly higher than those in OSBT,OSA and NOT groups(P<0.01).The positive rates of PI3K110α,p-AktSer473,β-Catenin and Ki-67 protein were statistically different between infiltrated and non-infiltrated specimens in OSC group(P<0.01),the positive rates of PI3K110α,p-AktSer473,β-Catenin and Ki-67 protein were statistically different between samples with and without lymph node metastasis in OSC group(P<0.01).Conclusion The loss of E-Cadherin,PTEN is associated with the over activation of PI3Kp110α,p-AktSer473,β-Catenin and Ki-67 proteins,which may be involved in the occurrence and development of OST,the detection of the above proteins may be used for the clinical diagnosis and prognosis of OST.
