基于关联规则和复杂系统熵聚类研究的视网膜静脉阻塞组方规律研究

Analysis on medication rule in prescriptions for retinal vein occlusion based on apriori and entropy clustering algorithm of complex system

目的探讨中医药方剂治疗视网膜静脉阻塞(RVO)的组方用药规律。方法搜集从各数据库建库—2018年在中国知网(CNKI)、万方数据库、维普数据库及PubMed数据库运用中医药治疗RVO的方剂,采用中医传承辅助平台(V2.5)软件,分析治疗RVO的组方用药规律。结果(1)通过筛选整理后共得出420条方剂,包含216味中药,其中用药频次≥10的中药共计75味。(2)共得出64种药物组合模式。其中,2味药的药对组合26条,3味药的核心组合22条,4味药及以上的核心组合模式16条;通过网络展示图,得出居于中心的药物有当归、赤芍、川芎、生地黄。(3)分析治疗RVO方剂中216味中药两两之间的关联度,得到关联系数>0.015的药对共37对;演化出3~5味药的核心组合,共计78个。(4)分析聚类得到新处方13首。结论应用中医传承辅助系统分析RVO的组方规律,揭示RVO的遣方用药以活血化瘀、行气为主,辨证加减施治,分析得出的药对组合和候选新方可为临床治疗和基础研究提供一定的线索与参考。

OBJECTIVE To analyze rule of Traditional Chinese Medicine prescriptions for retinal vein occlusion.METHODS Searching in CNKI,WANFANG,VIP,and PubMed database from the creation time to 2018 for literature about vein occlusion.The principles of frequency,pairing and grouping of different drugs were analyzed based on Apriori and entropy clustering algorithm of complex system by using Traditional Chinese Medicine inheritance support system(TCMISS V2.5).RESULTS(1)Four hundred and twenty prescriptions were analyzed.Totally,216 drugs were screened out.Seventy-five of them had frequency equal to or more than 10 times.(2)Sixty-four compound modes were discovered.Among them,the combinations which contained a pair of drugs were twenty-six.The number of the core combinations which contained 3 drugs were twenty two,while ones which contained 4 drugs were sixteen.The network graph showed that the core drugs were Chinese angelica,red peony root,Ligusticum wallichii,and root of rehmannia glutinosa.(3)The principles of frequency,pairing and grouping of the 216 drugs were analyzed.The drugs with correlation coefficient>0.015 were 37 pairs.The number of the core combinations which contained 3-5 drugs were seventy eight.(4)Thirteen new prescriptions were discovered.CONCLUSIONS TCMISS V2.5 helped analyze the rule of Traditional Chinese Medicine prescriptions for retinal vein occlusion,and it revealed that the treatment of retinal vein occlusion mainly included methods of promoting blood circulation,removing blood stasis and promoting qi,and modifications based on syndrome differentiation.The yielded new pairs of herbs and candidate formula could provide clues and references for clinical practice and basic research.
