全基因组预测哈茨木霉的效应子 被引量:2

Prediction of Candidate Effectors from Genome of Trichoderma harzianum

摘要 【目的】通过采用高通量技术筛选哈茨木霉菌株的效应子。【方法】根据哈茨木霉CBS 226.95菌株全基因组14065个蛋白序列信息,利用SignalP、TMHMM、TargetP和Protcomp等生物信息学软件和预测程序进行分泌蛋白预测。【结果】分析得到709个蛋白,占总蛋白的5.04%。再对709个分泌蛋白与胞外酶数据库进行比对分析、半胱氨酸含量、multiple tandem repeats分析,筛选获得<300个氨基酸的小分子蛋白作为候选效应蛋白。共获得24个蛋白,其中有3个是碳水化合物结合模块家族蛋白,其余21个为假定蛋白。【结论】本研究利用生物信息学方法从哈茨木霉基因组中预测出候选效应子,为进一步研究效应子在拮抗真菌与病原真菌及植物间的互作关系奠定了基础。 【Objective】Trichoderma harzianum is wildly used in biological control.Effectors played an important role in the interaction process.【Method】SignalP,TMHMM,TargetP and Protcomp bioinformatics methods were used to analyze the T.harzianum genome that contain 14065 protein.【Result】709 secreted proteins were obtained,accounting for 5.04%of the total protein.Based on the above secretory protein,the analysis with the extracellular enzyme database was done,then cysteine content and multiple tandem repeats were analyzed,and at last,the small molecule proteins with<300 amino acids were screened as candidate effector proteins.Finally,24 proteins were confirmed to be the effectors of T.harzianum.【Conclusion】In this study,candidate effectors were predicted from T.harzianum genome by bioinformatics method,which laid a foundation for further study on the interaction between antagonistic fungi,pathogenic fungi and plants.
