基于网络药理学的桃仁-红花药对治疗缺血性心脏病机制研究 被引量:23

Study on Mechanism of Drug Pair Persicae Semen-Carthami Flos in Treatment of Ischemic Heart Disease Based on Network Pharmacology

摘要 目的采用网络药理学方法研究活血化瘀药桃仁-红花药对治疗缺血性心脏病的作用机制。方法采用中药系统药理学数据库与分析平台(TCMSP)筛选药物成分及成分靶点,通过Genecards数据库进行疾病靶点预测,利用String平台进行蛋白相互作用(PPI)分析并构建PPI网络,应用Cytoscape3.6.1软件构建桃仁-红花药对治疗缺血性心脏病的成分-靶点-疾病网络,利用Bioconductor中R包clusterprofile version3.14.2进行GO功能注释和KEGG通路分析。结果筛选出桃仁-红花药对化学成分45个,作用靶点105个;PPI网络共有节点78个,边796条,平均度值20.41;GO分析条目共92个,包括血红素结合、细胞因子受体结合、过氧化酶活性、生长因子受体结合等;KEGG信号通路121条,包括白细胞介素-17信号通路、T细胞受体信号通路、血管内皮生长因子信号通路、肿瘤坏死因子信号通路、流体剪切应力和动脉粥样硬化信号通路、糖尿病并发症中的AGE-RAGE信号通路、雌激素信号通路、PI3K-Akt信号通路、MAPK信号通路、p53信号通路、核因子-κB信号通路、HIF-1信号通路等。结论桃仁-红花药对通过多靶点、多通路治疗缺血性心脏病,涉及的生物学途径可能包括炎症、血流动力学、氧化应激、细胞凋亡、信号传导等。 Objective To explore the mechanism of treating ischemic heart disease with the promoting blood circulation and reducing blood stasis drug pair Persicae Semen-Carthami Flos based on network pharmacology.Methods The chemical components and targets of Persicae Semen and Carthami Flos were screened by searching the TCMSP.Genecards database was used to predict disease targets,String platform was used to analyze protein-protein interactions(PPI),and PPI network was built.The component-target-disease network of Persicae Semen-Carthami Flos drug pair for treating ischemic heart disease was constructed using Cytoscape3.6.1 software.GO function annotation and KEGG pathway analysis were performed using R package clusterprofile version 3.14.2 in Bioconductor.Results A total of 45 chemical components and 105 targets were identified from Persicae Semen-Carthami Flos drug pair.There were 78 nodes and 796 edges in PPI network,and the average degree value was 20.41.Totally 92 GO analysis items were obtained,including heme binding,cytokine receptor binding,peroxidase activity,growth factor receptor binding,etc.;121 KEGG signaling pathways were obtained,including interleukin-17 signaling pathway,T cell receptor signaling pathway,VEGF signaling pathway,TNF signaling pathway,fluid shear stress and atherosclerotic signaling pathway,AGE-RAGE in diabetes complications signaling pathway,estrogen signaling pathway,PI3K-Akt signaling pathway,MAPK signaling pathway,p53 signaling pathway,NF-κB signaling pathway,HIF-1 signaling pathway,etc.Conclusion Persicae Semen-Carthami Flos drug pair treats ischemic heart disease through multiple targets and multiple pathways.The biological pathways involved may include inflammation,hemodynamics,oxidative stress,apoptosis,signal transmission,etc.
