光甘草定和氧化白藜芦醇协同治疗黄褐斑作用评价 被引量:3

摘要 采用体外酪氨酸酶抑制实验并结合响应曲面法优化了光甘草定和氧化白藜芦醇协同抑制酪氨酸酶配比;将光甘草定和氧化白藜芦醇按照较优活性配比制备成胶束溶液,中波紫外照射法复制小鼠黄褐斑模型,给药治疗后,检测小鼠血清和皮肤组织中的超氧化物歧化酶(SOD)及丙二醛(MDA)。结果表明,光甘草定和氧化白藜芦醇的最佳物质的量比为3∶5;光甘草定胶束溶液、氧化白藜芦醇胶束溶液及光甘草定和氧化白藜芦醇复配胶束溶液均可降低黄褐斑小鼠皮肤组织和血清中的脂质过氧化,提高SOD活性;与模型组相比,光甘草定和氧化白藜芦醇复配组在血清中的SOD活性升高了24.71U/m L,MDA含量降低了7.90U/m L;在皮肤组织中的SOD活性升高了15.14 U/mg prot,MDA含量降低了1.60 U/mg prot,说明光甘草定和氧化白藜芦醇复配胶束溶液效果最佳。光甘草定和氧化白藜芦醇对黄褐斑的形成具有保护作用,并且两者具有协同作用,联用效果更好。 The synergistic inhibition ratio of glabridin and oxyresveratrol to tyrosinase was optimized by in vitro tyrosinase inhibition experiments and response surface method.The micelle solution of glabridin and oxyresveratrol was prepared according to the optimum activity ratio,and the model mice with chloasma were replicated by medium-wave ultraviolet irradiation.After the micelle solution treatment,the superoxide dismutase(SOD)and malondialdehyde(MDA)in the serum and skin tissues of the mice were detected.The results showed that the optimal molar ratio of glabridin to oxyresveratrol was 3∶5.Glabridin micelle solution,oxyresveratrol micelle solution as well as glabridin and oxyresveratrol compound micelle solution all reduced lipid peroxidation in skin tissue and serum of mice with chloasma,and increased SOD activity.Compared with those of model group,SOD activity in serum of glabridin and oxyresveratrol compound group increased by 24.71 U/mL,MDA content decreased by 7.90 U/mL.SOD activity in skin tissue increased by 15.14 U/mg prot,MDA content decreased by 1.60 U/mg prot,indicating that glabridin and oxyresveratrol compound micelle solution had the best effect.Glabridin and oxyresveratrol have protective effect on the formation of chloasma,and the two have synergistic effect which works better when combined.
