摘要
目的:探讨毛兰素(Erianin,Eri)通过活化JNK通路诱导乳腺癌细胞凋亡的作用及分子机制。方法:通过MTT及克隆形成方法检测Eri对乳腺癌细胞增殖能力的影响作用;通过Hoechst染色及流式细胞术检测Eri对细胞凋亡的影响;通过Western blot检测Eri诱导细胞凋亡的作用以及JNK通路参与其中的机制。结果:Eri可以抑制乳腺癌细胞增殖,促进细胞凋亡,且Eri通过活化JNK/c-Jun信号通路发挥作用,促进凋亡发生。结论:Eri通过活化JNK/c-Jun信号通路,促进乳腺癌细胞凋亡,从而抑制细胞增殖。Eri可能成为治疗乳腺癌的新型候选药物。
Objective To investigate the role and molecular mechanism of Erianin(Eri)in inducing breast cancer cell apoptosis through activating JNK signaling pathway.Methods MTT and colony formation assay were used to assess the inhibitory effect of Eri on cell proliferation in human breast cancer cells.The effect of Eri on cell apoptosis was detected by Ho⁃echst staining and flow cytometry.The Eri-induced apoptosis and the mechanism of JNK pathway were detected by western blot.Results Eri inhibited proliferation and induced apoptosis in breast cancer cells.Eri promoted apoptosis via activation of JNK/c-Jun signaling pathway.Conclusion Eri can promote the apoptosis of breast cancer cells by activating JNK/c-Jun signaling pathway and inhibit cell proliferation.Eri may become a new drug candidate for the treatment of breast cancer.
