星点设计-效应面法优化武当山白芍中芍药苷的提取工艺研究 被引量:4

Optimization of Extractive Technology of Paeoniflorin from Radix Paeoniae Alba in Wudang Mountains by Central Composite Design-Response Surface Method

摘要 目的:优化武当山白芍中芍药苷的提取工艺,采用HPLC法测定不同年限白芍中芍药苷的含量,旨在优化芍药苷的提取工艺,为地方企业规范化种植和采收提供一定的参考。方法:采用HPLC法,利用星点设计-效应面法优化芍药苷的提取工艺,并采用优化后工艺测定2~5年生白芍中芍药苷的含量。结果:芍药苷的最佳提取工艺为:以30%乙醇为提取溶剂,料液比为1∶52,超声提取10 min。随着白芍生长年限的增加,芍药苷的含量并未呈现明显的递增趋势。结论:采用星点设计-效应面法优化所得芍药苷的最佳提取工艺,方法简单、可靠,时间短、能耗低,提取率高。芍药苷的含量并未出现随着生长年限延长明显递增的趋势,从经济学角度出发,建议白芍采挖年限不宜过长。 Objective To optimize the extraction process of paeoniflorin from Radix paeoniae Alba in Wudang mountains,and determine the content of paeoniflorin in Radix Paeoniae Alba of different ages by HPLC,so as to optimize the extraction process of paeoniflorin,and provide certain reference for local enterprises to standardize planting and harvesting.Methods HPLC method was used to optimize the extraction process of paeoniflorin by central composite design-response surface method,and the content of paeoniflorin with 2~5 years old were determined by the optimized process.Results The optimal extraction process of paeoniflorin was as follows:30% ethanol as extraction solvent,solid-liquid ratio of 1∶52,ultrasonic extraction for 10 min.The content of paeoniflorin did not show an obvious increasing trend with the growth years of Radix paeonia Alba.Conclusion The optimum extraction process of paeoniflorin optimized by central composite design response surface methodology was simple,reliable,short time,low energy consumption and high extraction rate.The content of paeoniflorin did not increase significantly with the extension of growth years.From the perspective of economics,it is suggested that the mining period of Radix paeonia Alba should not be too long.
