摘要
目的探讨阿尔茨海默病(AD)大鼠模型采用电针治疗对学习记忆能力、海马组织中谷氨酸受体相互作用蛋白1(GRIP1)、GRIP2及血清中基质细胞衍生因子-1(SDF-1)、趋化因子单核细胞趋化蛋白1(MCP-1)水平的影响。方法选取成年雄性SD大鼠48只(SPF级),随机分为空白组、假手术组、模型组、实验组(双侧肾俞、百会穴进行电针治疗,1次/天,连续治疗2周),采用海马CA1区注射寡聚态Aβ25-35制备模型。采用Morri水迷宫实验检测各组大鼠的行为学改变,采用免疫组化染色检测各组大鼠海马组织中GRIP1、GRIP2蛋白,采用ELISA法检测各组大鼠的血清MCP-1、SDF-1水平。结果模型组大鼠的平台搜寻时间显著的长于空白组和假手术组(P<0.05),模型组大鼠的第二象限活动时间、跨越平台次数均显著的低于空白组和假手术组(P<0.05);实验组的平台搜寻时间显著低于空白组(P<0.05),实验组的第二象限活动时间、跨越平台次数显著高于空白组(P<0.05);模型组大鼠的血清MCP-1水平高于空白组和假手术组(P<0.05),模型组大鼠的血清SDF-1水平显著的低于空白组和假手术组(P<0.05);实验组的血清MCP-1水平低于空白组(P<0.05),实验组的血清SDF-1水平高于空白组(P<0.05)。结论电针治疗AD大鼠能显著的改善大鼠的学习记忆能力,与提高海马组织中GRIP1、GRIP2蛋白、血清SDF-1水平及降低MCP-1水平有关。
Objective Effect of electroacupuncture for treatment of Alzheimer's disease(AD)in the hippocampus of rats learning and memory ability,glutamate receptor interacting protein 1(GRIP1),GRIP2 and serum chemokine monocyte chemotactic protein 1(MCP-1)and stromal cell-derived factor-1(SDF-1)level of influence.Methods Forty-eight adult SD male rats(SPF grade),were randomly divided into control group,sham operation group,model group,experimental group(bilateral Shenshu and Baihui electroacupuncture treatment,1 times/day,continuous treatment for 2 weeks),poly beta 25-35 state A model was prepared by injection of oligonucleotides in hippocampal CA1 region.Morri water maze test was used to detect behavioral changes in each group.Immunohistochemical staining was used to detect GRIP1 and GRIP2 protein in hippocampus of rats in each group.Serum levels of MCP-1 and SDF-1 were detected by ELISA.Results The rats in the model group was significantly longer than the time the search platform for control group and sham operation group(P<0.05),the rats in the model group the second quadrant time and times of crossing the platform were significantly lower than the control group and sham operation group(P<0.05);the experimental group was significantly lower than that of blank time search platform(P<0.05),experimental group a group of second quadrant time and times of crossing the platform was significantly higher than that of control group(P<0.05);the level of serum MCP-1 of the rats in the model group was higher than that of control group and sham operation group(P<0.05),model group rats serum SDF-1 level was significantly lower than that of control group and sham operation group(P<0.05);the serum MCP-1 level of the experimental group lower than that of the control group(P<0.05),serum SDF-1 level of experimental group was higher than control group(P<0.05).Conclusion Electro acupuncture treatment of AD rats can significantly improve learning and memory ability in rats,which is related to the increase of GRIP1,GRIP2 protein,serum SDF-1 level and MCP-1 level in hippocampus.
