摘要
目的探讨颗粒细胞表面间隙连接蛋白(connexin,Cx)37的表达与卵泡成熟情况及辅助生殖技术(IVF-ET)妊娠结局的相关性。方法选择64例拟行常规IVF-ET治疗患者的卵泡液,梯度密度离心后收集颗粒细胞,采用流式细胞术检测颗粒细胞表面Cx37的表达,与MⅡ卵率、hCG日孕酮及雌二醇值进行相关性研究;根据术后14天hCG值分为妊娠组与未妊娠组,比较两组间患者年龄、基础内分泌值、获卵数、Cx37表达情况。结果妊娠组与非妊娠组被检者的年龄、基础内分泌值、hCG日孕酮及雌二醇比较,差异均无统计学意义(P>0.05);妊娠组MⅡ卵率高于非妊娠组MⅡ卵率,差异有统计学意义(P<0.05);妊娠组Cx37的表达低于非妊娠组,差异具有统计学意义(P<0.05);Cx37的表达水平与MⅡ卵率呈负相关性;Cx37的表达水平与hCG日孕酮、雌二醇水平呈正相关性,差异均有统计学意义(P<0.05)。结论推测将流式细胞术推广用于临床定量测定Cx37,在一定程度可对卵泡成熟做出预判,并以期预测辅助生殖技术助孕的妊娠率。
Objective To investigate the relationship of connexin 37 in granulosa cells with follicular maturation and IVF-ET pregnancy outcome.Methods 64 patients who were to be treated with IVF-ET were studied.Granulosa cells were collected by used density gradient centrifugation.Flow cytometry was used to detect the expression of Cx37 on the surface of granulosa cells.Comparing MII follicles ratio,progesterone and estrogen level of hCG day.Dividing patients into pregnant group and non-pregnant group according to hCG results after embryo transfer.Comparing age,basic estrogen and progesterone,number of retrieved oocytes and expression of Cx37.Results Age,basic level of estrogen and progesterone,number of retrieved that There were no statistical significantly differences between pregnant group with non-pregnant group(P>0.05).The MII oocyte ration of pregnant group was higher than non-pregnant group(P<0.05).The expression of Cx37 in pregnancy group was lower than that in non-pregnancy group(P<0.05).The results showed that there were a negative correlation between the expression of Cx37 and the egg rate of MII(P<0.05).There were a positive correlation between the expression level of Cx37 with progesterone and estrogen level of hCG day(P<0.05).Conclusion It is speculated that flow cytometry can be used in clinical quantitative determination of Cx37,which can predict the follicular maturation to a certain extent,and predict the pregnancy rate of assisted reproductive technology.
