经方吴茱萸汤对偏头痛模型大鼠的预防性治疗作用研究

Study on Preventive and Therapeutic Effects of Wuzhuyu Decoction(吴茱萸汤)on Migraine Model Rats

目的:观察经方吴茱萸汤对偏头痛模型大鼠行为学、血浆降钙素基因相关肽(CGRP)、P物质(SP)含量的影响,探讨吴茱萸汤对偏头痛预防性治疗作用的可能机制。方法:60只Wistar大鼠随机分成6组:正常组、模型组、西比灵组、吴茱萸汤高剂量组、吴茱萸汤中剂量组和吴茱萸汤低剂量组。连续灌胃药物7 d,末次给药30 min后除正常组外其它各组大鼠按10 mg/kg皮下注射硝酸甘油制备偏头痛模型,正常组注射生理盐水。录像记录并观察大鼠行为学表现后,用酶联免疫吸附法(ELISA)检测血浆CGRP、SP含量。结果:与正常组比较,模型组、西比灵组、吴茱萸汤高剂量组、吴茱萸汤中剂量组、吴茱萸汤低剂量组大鼠均出现耳红、频繁挠头,血浆CGRP和SP含量升高(P<0.05)。与模型组比较,西比灵组、吴茱萸汤高剂量组、吴茱萸汤中剂量组、吴茱萸汤低剂量组大鼠耳红持续时间缩短,挠头次数减少(P<0.05),血浆CGRP、SP含量明显降低(P<0.05)。结论:吴茱萸汤可能通过调节血浆CGRP、SP的含量改善大鼠偏头痛症状,对偏头痛具有预防性治疗作用。

Objective:To observe the effects of Wuzhuyu Decoction on behavior,contents of plasma calcitonin gene-related peptide(CGRP)and substance P(SP)in migraine model rats,and to explore the possible mechanism of Wuzhuyu Decoction in preventive treatment of migraine.Methods:60 Wistar rats were randomly divided into 6 groups:normal group,model group,Sibelium group and group with high dose of Wuzhuyu Decoction,group with medium dose of Wuzhuyu Decoction and group with low dose of Wuzhuyu Decoction.After continuous gavage for 7d and 30 min after the last administration,the migraine model was made.In addition to the normal group,the other groups were injected subcutaneously with nitroglycerin injection(10 mg/kg),and the normal group was injected with normal saline.After recording and observing the behavior of rats,the contents of CGRP and SP in plasma were determined by enzyme linked immunosorbent assay(ELISA).Results:Compared with the normal group,the rats in the model group,Sibelium group and Wuzhuyu Decoction groups with high,medium and low dose all showed red ears and frequent head scratching,and the levels of plasma CGRP and SP were increased(P<0.05).Compared with the model group,the duration of ear red was shortened,the number of scratching head was decreased(P<0.05),and the content of CGRP and SP in plasma was significantly decreased(P<0.05)in Sibelium group and the high,medium and low dose groups of Wuzhuyu Decoction.Conclusion:Wuzhuyu Decoction may improve the symptom of migraine in rats by regulating the content of plasma CGRP and SP,so as to achieve the preventive treatment of migraine.
