bFGF转染骨髓间充质干细胞移植对新生骨成熟过程的生物力学影响——兔胫骨延长模型研究 被引量:2

摘要 目的探讨碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)基因转染的骨髓间充质干细胞移植对家兔胫骨截骨延长模型中新生骨成熟过程的生物力学影响的实验研究。方法采用随机区组设计,将48只纯种新西兰实验用大白兔按体重相同或相近分为16个区组。建立兔胫骨截骨延长模型,延长结束后,实验组于牵张间隙内注入bFGF转染的骨髓间充质干细胞,对照组于牵张间隙内注入骨髓间充质干细胞,空白组不注入任何物质。结果在固定期4周和8周,实验组牵张区在X线片结果、HE染色观察方面成骨质量明显优于对照组和空白组。12周后取各组牵张成骨区标本做骨密度检测以及生物力学测定,结果与对照组、空白组相比,实验组有明显优势。结论bFGF基因转染的骨髓间充质干细胞移植对兔胫骨延长区新生骨成熟过程有明显的促进作用。 Objective To investigate the biomechanical effects of bone marrow mesenchymal stem cell transplantation transfected with basic fibroblast growth factor gene on the maturation process of newborn bone in tibia osteogenesis model of rabbits.Methods Using a random block design,48 purebred New Zealand experimental white rabbits were divided into 16 blocks according to the same or similar body weight.A rabbit tibia osteotomy extension model was established.After the extension,the experimental group was injected with bFGF-transfected bone marrow mesenchymal stem cells in the distraction space.The control group was injected with bone marrow mesenchymal stem cells in the distraction space.Results In the fixed period of 4 weeks and 8 weeks,the osteogenesis quality of the experimental group in the distracted area was significantly better than that of the control group and the blank group in terms of radiograph results and HE staining observation.After 12 weeks,samples of distraction osteogenesis in each group were taken for bone density determination and biomechanical test.The results showed that the above two indicators of the experimental group were significantly better than those of the control group and the blank group.Conclusion The transplantation of bFGF gene-transfected bone marrow mesenchymal stem cells can significantly promote the maturation of new bone in the tibia extension area of rabbits.
