摘要
目的建立仙游青黛HPLC指纹图谱和简化靛蓝含量测定。方法采用C18柱(4.6mm×250mm,5μm);甲醇-水(70∶30)洗脱;流速1.0mL·min^-1;检测波长:292nm,进样量:10μL,测定了11批青黛样品。结果相同或不同产家的不同批号的11批青黛指纹图谱之间的相似度均大于0.99,靛蓝按上述色谱条件下测定的含量与药典法测定的含量差异甚微。结论所建立的青黛HPLC指纹图谱为仙游各生产企业或作坊的青黛质量提供参考,并统一了靛蓝和靛玉红测定时的色谱条件和提取溶剂。
OBJECTIVE To establish the HPLC fingerprint chromatogram of Xianyou indigo naturalis and simplify the determination of indigo.METHODS The analysis was achieved with a C18 column(4.6 mm×250 mm,5μm)by 70%(v/v)methanol in water.The flow rate was 1.0 mL·min^-1 with wavelength:λ=292 nm.Its injection volume was 10μL.11 samples were analyzed by the above method.RESULTS The similarity of indigo naturalis was above 0.99 within the products by the same manufacture and the different manufacture.There was little difference between the content of Indigo determined under the above chromatographic conditions and that determined by Pharmacopoeia method.CONCLUSION The established HPLC fingerprint can be used to provide a reference for the quality evaluation of indigo naturalis from different manufacture in Xianyou.The chromatographic conditions and extraction solvents for the determination of indigo and Indirubin were unified.
