摘要
目的:观察CD_3AK抗CD3单克隆抗体)的活化及对肿瘤细胞的杀伤活性。方法:以不同的肿瘤细胞为靶细胞,以人外周血淋巴细胞为效应细胞,在体外用不同浓度的抗CD3单克隆抗体和/或IL-2进行激活,用MTT法观察细胞毒杀伤作用。结果:CD3AK的扩增率较LAK高一个数量级,且对不同的肿瘤细胞均有杀伤作用。其18h杀伤均大于4h。结论:由于CD3AK较LAK对IL-2的需要量少,扩增率高,而比LAK更有临床应用发展前景。
Objective: To study the activation, and the cytotoxity to tumor cell lines, of CD3AK. Methods:Several tumor cell lines were chosen as targt cells and human Peripheral blood lymphocytes (PBL) as effect cells. The PBL was Activated outside with CD3 McAb and /or IL - 2 at different concentrations. And then the cytotokity of effect cells was observed to apt cells by MTT. Results: The propagation rate of CD3AN is much higher than tha of ase- CD3AK. Furthermore, it can destroy tumor cells of various types, with an 18 - h cytotoxity stronger than the 3 - h one. An: The clinic applieation of CD3AK might be more promising beause, compared with LAK, it has a higher increase rate and a fowr dependence on IL - 2.
