摘要
(1)目的调查女性阴道内正常菌群分布,筛选阴道微生态制剂种子菌。(2)方法取健康体检女性阴道分泌物经镜检及五联卡检测后细菌培养,挑选优势菌落革兰染色后生化反应初步诊断,采用实时荧光定量PCR进行分子生物学鉴定;直方图描述阴道正常菌群频数分布,筛选种子菌。(3)结果 224份健康体检女性阴道分泌物,共分离并鉴定7种(392株)细菌,其中嗜酸乳杆菌123株,分离率54.9%(123/224);发酵乳杆菌98株,分离率43.8%(98/224);棒状杆菌59株,分离率26.3%(59/224);缓症链球菌37株,分离率16.5%(37/224);表皮葡萄球菌29株,分离率12.9%(29/224);马胃葡萄球菌28株,分离率12.5%(28/224);粪肠球菌19株,分离率8.5%(19/224)。(4)结论健康女性阴道菌群的优势菌为发酵乳杆菌、嗜酸乳杆菌及棒状杆菌,可作为阴道微生态制剂的候选种子菌株。
Objective To investigate the distribution of normal flora in female vagina,and to screen the seed bacteria of vaginal microecological agents.Methods Bacterial culture for vaginal secretions from healthy women was carried out after microscopic examination and quintuple examination.The dominant colonies were selected for preliminary diagnosis by biochemical reaction as well as gram staining.Thereafter,molecular biological identification was conducted with real-time fluorescence quantitative polymerase chain reaction.The candidate seed strains of vagina micro-ecological preparation were screened based on the frequency distribution of normal vaginal flora described by histogram.Results 7 kinds of bacteria were isolated and identified in 224 healthy women,including 123 strains of lactobacillus acidophilus and with an isolation rate of 54.9%(123/224),98 strains of lactobacillus fermentans and with 43.8%(98/224),59 strains of corynebacterium with 26.3%(59/224),37 strains of streptococcus mitis with 16.5%(37/224),29 strains of staphylococcus epidermidis with 12.9%(29/224),28 strains of staphylococcus EQUIS with 12.5%(28/224),and 19 strains of enterococcus faecalis with 8.5%(19/224).Conclusion Lactobacillus fermentans,lactobacillus acidophilus and corynebacterium are the dominant bacteria in vagina flora of healthy women,therefor,they can be served as candidate seed strains for developing vagina micro-ecological preparation.
