马齿苋多糖对荷宫颈癌裸鼠肿瘤生长的影响 被引量:6

Effects of purslane polysaccharides on tumor growth in nude mice with cervical cancer

摘要 目的探讨马齿苋多糖对荷宫颈癌裸鼠肿瘤生长及移植瘤组织增殖细胞核抗原(PCNA)、Ethera go-go 1(Eag1)表达的影响。方法用裸鼠左侧腋窝外侧皮下接种宫颈癌Caski细胞的方法构建荷宫颈癌裸鼠模型。将成功构建的荷宫颈癌裸鼠随机分为模型组及低、高剂量实验组,各12只。低、高剂量实验组裸鼠分别予以灌胃马齿苋多糖100,200 mg·kg-1,每日1次;模型组予以灌胃等量生理盐水,连续给药14 d。测定抑瘤率及胸腺指数、脾脏指数;用蛋白质印迹(Western blot)法检测移植瘤组织中PCNA、Eag1蛋白的表达水平。结果低、高剂量实验组抑瘤率分别为(9.58±3.64)%和(45.61±4.32)%;模型组、低、高剂量实验组裸鼠胸腺指数分别为(28.21±3.87),(39.42±13.63)和(57.65±9.38)mg·10 g^-1;脾脏指数分别为(98.66±11.74),(156.02±19.15)和(178.49±10.83)mg·10 g^-1;PCNA蛋白相对表达量分别为0.81±0.14,0.63±0.11,0.42±0.08;Eag1蛋白相对表达量分别为0.69±0.13,0.52±0.09,0.40±0.06。上述指标,低、高剂量实验组分别与模型组比较(抑瘤率除外),低、高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论马齿苋多糖可有效抑制荷宫颈癌裸鼠肿瘤的生长,促进移植瘤细胞凋亡,同时改善免疫功能,其机制可能与抑制移植瘤组织PCNA、Eag1蛋白表达相关。 Objective To investigate the effects of purslane polysaccharides on tumor growth and expression of proliferating cell nuclear antigen(PCNA)and Etherà go-go 1(Eag1)in xenograft tissues in nude mice with cervical cancer.Methods The nude mice models with cervical cancer were constructed by subcutaneously inoculating cervical cancer Caski cells on the left lateral armpit of nude mice.The successfully constructed nude mice bearing cervical cancer were randomly assigned to the model group and the exp-L/-H group,with 12 mice in each group.The nude mice in the exp-L/-H groups were gavaged with 100,200 mg·kg^-1 of purslane polysaccharides once daily,and the model group were gavaged with an equal amount of normal saline for 14 d.The tumor inhibition rate,thymus index and spleen index were measured;Western blot was used to detect the expression levels of PCNA and Eag1 protein in the xenograft tissues.Results The tumor inhibition rates of the exp-L/-H groups were(9.58±3.64),(45.61±4.32)%;the thymus indexes of the model group and exp-L/-H group were(28.21±3.87),(39.42±13.63)and(57.65±9.38)mg·10 g^-1;the spleen indexes were(98.66±11.74),(156.02±19.15)and(178.49±10.83)mg·10 g^-1;the relative expression levels of PCNA protein were 0.81±0.14,0.63±0.11,0.42±0.08;the relative expression levels of Eag1 protein were 0.69±0.13,0.52±0.09,0.40±0.06.There were statistically significant differences between exp-L/-H group and model group in the above factors(except tumor inhibition rate),and between exp-L group and exp-H group in the above factors(all P<0.05).Conclusion Purslane polysaccharides can effectively inhibit the tumor growth of cervical cancer-bearing nude mice,promote apoptosis of xenograft cells,and improve immune function,the mechanism may be related to the inhibition of PCNA and Eag1 protein expression in xenograft tissues.
