麦瓶草谷胱甘肽体外抗氧化活性研究 被引量:2

摘要 实验主要研究麦瓶草谷胱甘肽(GSH)体外的抗氧化活性,采用超声波提取法,以提高谷胱甘肽溶出率。通过实验考察,得到麦瓶草谷胱甘肽的较优提取条件:提取温度室温、乙醇浓度40%、料液比1:4.5 g/mL、提取时间2 h、转速12000 r/min、离心时间15 min、提取率32.374%,得到麦瓶草谷胱甘肽提取液后,通过真空干燥,最终得到干燥的麦瓶草谷胱甘肽提取物。将麦瓶草谷胱甘肽提取物进行不同程度的稀释,采用紫外分光光度法对麦瓶草谷胱甘肽稀释液进行谷胱甘肽含量测定。通过将麦瓶草内提取出来的谷胱甘肽提取物对金属离子Fe^2+螯合能力的测定、对羟基自由基清除能力的测定、对DPPH自由基清除能力的测定、对超氧阴离子自由基的测定来研究其在体外的抗氧化活性。当谷胱甘肽提取物稀释后质量浓度60μg/mL时,对金属离子Fe^2+螯合率达到了58.056%;当谷胱甘肽提取物稀释后质量浓度为20μg/mL时,对羟基自由基的清除率达49.383%,当质量浓度60μg/mL时,清除率达55.556%;当谷胱甘肽提取物稀释后质量浓度为20μg/mL时,对超氧阴离子自由基清除率达90.521%,当质量浓度60μg/mL时,清除率达99.065%,清除能力很强。实验结果表明,麦瓶草谷胱甘肽具有较好的体外抗氧化活性。为麦瓶草的进一步开发利用提供了理论参考。 It was to study the antioxidant activity of glutathione(GSH)in Silene conoidea Lin vitro.Ultrasonic extraction method was used to improve the dissolution rate of glutathione.The better extraction conditions of glutathione were obtained by experimental investigation.The extraction temperature was room temperature.The ethanol concentration was 40%.The feed-liquid ratio was 1:4.5 g/mL.The extraction time was 2 h and the rotational speed was 12000 r·min-1.The centrifugation time was 15 min and the extraction rate was 32.374%.After obtaining the extract of glutamine,the dried glutamine extract was finally obtained by vacuum drying.The content of glutathione was determined by UV spectrophotometry.The antioxidant activity of glutathione extracted from silene conoidea L.was studied by determining the chelatingability of metalion Fe^2+,scavenging hydroxyl radical,scavenging DPPH radical and scavenging superoxide anion radical in vitro.When the mass concentration was diluted to 60μg/m L,the chelating rate of mental ions Fe^2+reached 58.056%.When the mass concentration was diluted to 20μg/mL,the scavening rate of hydroxyl radical reached 49.383%.When the mass concentration was diluted to 60μg/m L,the scavening rate of hydroxyl radical reached 55.556%.When the mass concentration was diluted to 20μg/m L,the scavening rate of superoxide anion radical reached 90.521%.When the mass concentration was 60μg/m L,the scavening rate of superoxide anion radicalreached 99.065%.The results showed that glutathione had good anti-oxidant activity in vitro.It provides a theoretical reference for further development and utilization of Silene conoidea L.
