雌性小鼠不同时期雌激素及其受体表达对Th1/Th2和Th17/Treg免疫平衡的影响 被引量:3

Effects of estrogen and its receptor expression on the immune balance of Th1/Th2 and Th17/Treg in female mice at different stages

摘要 目的探索雌性小鼠不同时期雌激素及其受体表达对Th1/Th2和Th17/Treg免疫平衡的影响。方法取2月龄雌性C57BL/6小鼠随机分为发情间期组、发情期组、妊娠期组,取13月龄雌性小鼠作为绝经期组,每组10只。分别检测4组小鼠血清雌激素(E2)水平、脾脏雌激素受体α(ERα)、雌激素受体β(ERβ)mRNA水平以及脾脏单个核细胞Th1、Th2、Th17、Treg亚群比例,计算Th1/Th2、Th17/Treg比值,并分析E2水平、ERαmRNA水平分别与Th1/Th2、Th17/Treg比值的相关性。结果与发情间期组比,发情期组、妊娠期组小鼠E2水平、ERαmRNA水平、Th2、Treg亚群比例升高(P<0.05),Th1、Th17亚群比例、Th1/Th2、Th17/Treg比值降低(P<0.05);绝经期组小鼠E2水平、ERαmRNA水平、Th2、Treg亚群比例降低(P<0.05),Th1、Th17亚群比例、Th1/Th2、Th17/Treg比值升高(P<0.05)。E2水平、ERαmRNA水平分别与Th1/Th2、Th17/Treg比值呈负相关(P<0.05)。结论雌性小鼠不同时期雌激素及其受体表达水平不同,且与Th1/Th2、Th17/Treg比值呈负相关,可能调节Th1/Th2和Th17/Treg免疫平衡。 Objective To explore the effects of estrogen and its receptor expression at different stages on the immune balance of Th1/Th2 and Th17/Treg in female mice.Methods Female C57BL/6 mice aged 2 months were randomly divided into estrus interval group(control group),estrus group and pregnancy group,and female mice aged 13 months were selected as menopause group,with 10 mice in each group.Serum estrogen(E2)levels,the mRNA levels of spleen estrogen receptorα(ERα),estrogen receptorβ(ERβ)levels and the proportion of Th1,Th2,Th17 and Treg subgroups in spleen mononuclear cells were detected in four groups of mice respectively,the Th1/Th2 and Th17/Treg values were calculated,and the correlations of E2 level and ERαmRNA level with Th1/Th2 and Th17/Treg values were analyzed.Results Compared with the estrus interval group,the E2 level,ERαmRNA level,Th2 and Treg subsets proportions of mice in estrus group and pregnancy group increased(P<0.05),Th1 and Th17 subsets proportions,Th1/Th2,Th17/Treg values decreased(P<0.05).The levels of E2,ERαmRNA,proportions of Th2 and Treg subsets decreased in the menopausal group(P<0.05),and the proportions of Th1 and Th17 subsets,Th1/Th2 and Th17/Treg values increased(P<0.05).The levels of E2 and ERαmRNA were negatively correlated with Th1/Th2 and Th17/Treg values respectively(P<0.05).Conclusion The expression levels of estrogen and its receptors in female mice at different stages are different,and are negatively correlated with Th1/Th2 and Th17/Treg values,which may regulate the immune balance of Th1/Th2 and Th17/Treg.
