LC-MS分析5味山姜属中药不同极性部位的共有成分研究

Analysis of Common Components in Different Parts of Five Alpinia Herbs by LC-MS

目的建立LC-MS方法研究5味山姜属中药不同极性部位的化学成分,分析化学成分的差异,找出5味中药不同极性部位的共有成分。方法色谱条件:不同极性部位采用不同条件,质谱条件:ESI离子源;正、负离子扫描模式;质量扫描范围:m/z 100~1200 Da。研究5味山姜属中药共性化学成分,比较成分差异性,分析亲缘关系与化学成分的相关性。结果5味山姜属中药不同提取部位的化学成分差异较大,但也存在共有成分,其中石油醚、乙酸乙酯、正丁醇、水部位分别比对出2、4、5、3个共有成分;其中标定出乙酸乙酯部位共有成分为原儿茶酸、原儿茶醛、4-香豆酸,正丁醇部位共有成分为原儿茶酸、柠檬酸,水部位共有成分为柠檬酸。结论5味药不同极性部位化学成分差异较大,存在共有成分,这些共有的成分可能与其辛温药性-温中散寒功效有关,为探讨山姜属中药亲缘关系与化学成分相关性提供了思路。

Objective To establish LC-MS conditions to analyze the common chemical components of different parts of five Alpinia herbs and analyze the difference.Methods The different chromatographic conditions were used for different parts of five Alpinia herbs.The MS conditions:ESI ion source;positive and negative ion scanning mode;mass scanning range:m/z 100 to 1200 Da.We studied the common chemical components of five Alpinia herbs,and compared the differences.The correlation between kinship and chemical components were analyzed.Results The chemical components of different parts of five Alpinia herbs were quite different,but there were also common components.The LC-MS analysis showed 2,4,5,and 3 common components in petroleum ether,ethyl acetate,n-butanol and water parts.Among them,the common components were the ethyl acetate part contained protocatechuic acid,protocatechuic aldehyde,4-coumaric acid,the n-butanol part contained protocatechuic acid and citric acid.The water part contained citric acid.Conclusion It is found that there are common components in different parts of five Alpinia herbs,but the chemical components are also different.These common components may be related to their pungent properties-warming and dispelling cold effect.It provides a way of thinking about the relationship between the kinship and chemical components of Alpinia herbs.
