培养基类型及贮藏温度对文冠果花粉离体培养萌发效果的影响分析

Effects of Medium Type and Storage Temperature on Germinatior of Xanthoceras sorbifolia Pollen in Vitro Culture

在文冠果杂交育种的过程中,一个至关重要的因素即为花粉的萌发活力,为了对花粉活力的影响因素进行研究,特开展了不同培养基类型、贮藏温度对文冠果花粉离体培养萌发效果的探究,结果表明,蔗糖100 g·L^-1、硼酸0.1 g·L^-1、硝酸钙0.25 g·L^-1、pH值6.0的培养基处理下文冠果花粉萌发率为90.7%,效果最好,在贮藏7 d的情况下,贮藏温度-80℃下萌发率为87.3%,效果最佳。

In the process of crossbreeding of Xanthoceras sorbifolia,a vital factor is the germination vigor of pollen.In order to study the influencing factors of pollen vigor,different media types and storage temperature have been specially developed for Xanthoceras sorbifolium odorifera The research on the germination effect of in vitro culture showed that 100 g·L^-1 of sucrose,0.1 g·L^-1 of boric acid,0.25 g·L^-1 of calcium nitrate,and a pH value of 6.0 had the best germination rate of 90.7%,and the effect was the best.Under the condition of 7 days storage,the germination rate was 87.3%at the storage temperature of-80℃,and the effect was the best.
