沉默OIP5对肺腺癌A549细胞迁移侵袭的影响研究 被引量:1

摘要 目的探讨Opa相互作用蛋白5(Opa interacting protein 5,OIP5)对肺腺癌A549细胞迁移和侵袭能力的影响。方法利用RNA干扰技术,沉默肺腺癌A549细胞中OIP5表达后,通过蛋白质印迹法检测OIP5的表达效率;通过transwell小室迁移及侵袭实验,分别检测OIP5沉默对肺腺癌A549细胞迁移和侵袭能力的影响。通过蛋白质印迹法,检测上皮标记物E钙黏蛋白(E-cadherin)和间质标记物波形蛋白(Vimentin)的表达,评价上皮间质转化(Epithelial-mesenchymal transition,EMT)过程。结果沉默OIP5后肺腺癌A549细胞OIP5蛋白表达水平明显降低(t=11.862,P<0.05),迁移、侵袭能力明显减弱(F=44.168、37.883,P值均<0.05),E-cadherin蛋白表达增加(F=39.787,P<0.05),Vimentin蛋白表达减少(F=40.164,P<0.05)。结论靶向沉默OIP5抑制肺腺癌A549侵袭、迁移能力,其作用机制可能是通过抑制细胞EMT来实现的。 Objective To explore the effect of Opa interaction protein 5(OIP5)on lung adenocarcinoma A549 cell migration and invasion ability.Methods After silencing the expression of OIP5 in A549 cells of lung adenocarcinoma with RNA interference technology,the expression efficiency of OIP5 was detected by Western blot.The effects of OIP5 silencing on migration and invasion of lung adenocarcinoma A549 cells were detected by transwell assay.The expression of Epithelial markers E-cadherin and Vimentin was detected by Western blot to evaluate the epithelial-mesenchymal transition(EMT).Results After OIP5 silencing,OIP5 protein expression level in A549 cells of lung adenocarcinoma was significantly decreased(t=11.862,P<0.05),migration and invasion ability were significantly decreased(F=44.168,37.883,all P<0.05),E-cadherin protein expression was increased(F=39.787,P<0.05),and Vimentin protein expression was decreased(F=40.164,P<0.05).Conclusion Targeted silencing OIP5 can inhibit the invasion and migration of lung adenocarcinoma A549,possibly by inhibiting EMT.
