摘要
目的:探讨氧化苦参碱(OMT)对胶原诱导性关节炎(CIA)小鼠滤泡辅助性T(Tfh)细胞比例和功能的影响。方法:构建牛Ⅱ型胶原和完全弗氏佐剂诱导的DBA/1J CIA小鼠模型,于首次免疫后第15天腹腔注射不同剂量的OMT,持续30 d。监测CIA小鼠的关节肿胀程度及关节炎评分;HE染色观察小鼠踝关节组织病理变化;提取脾细胞用流式细胞术检测Tfh细胞比例;采集小鼠眼球血,ELISA法测定小鼠血清Ⅱ型胶原抗体(CⅡ-Ab)和IL-21含量。结果:OMT可明显缓解CIA小鼠足爪肿胀,降低关节炎指数,减轻关节滑膜增生和炎症细胞浸润,降低脾Tfh细胞比例,抑制血清IL-21和CⅡ-Ab表达。结论:OMT可能通过调节Tfh细胞缓解RA。
Objective:To investigate effect of oxymatrine(OMT) on frequency and function of T follicular helper(Tfh) cells in mice with collagen-induced arthritis(CIA).Methods:DBA/1 J mice model were constructed by type Ⅱ collagen and CFA.15 d after a primary immunization,mice were treated with different concentrations of OMT i.p for 30 days.Paw swelling and arthritic score were evaluated.HE staining observed joint histopathology.Percentage of Tfh cells in mouse spleens by flow cytometry.Collected blood from eyes of mice.Expression levels of IL-21 and collagen type Ⅱ antibody(CⅡ-Ab) were measured by ELISA.Results:OMT significantly relieved paw swelling,reduced arthritic score and alleviated synovial hyperplasia in CIA mice.Percentage of Tfh cells were markedly down-regulated by OMT,serum levels of IL-21 and CⅡ-Ab were decreased.Conclusion:OMT exhibited a protective effect on RA via regulating percentage and function of Tfh cells.
