TBX3 affects proliferation,invasion and migration of gastric cancer cells through Wnt/β-catenin signaling pathway

摘要 目的:探讨TBX3对胃癌细胞增殖、侵袭、迁移和Wnt/β-catenin信号通路的影响,阐明TBX3在胃癌中的可能作用机制。方法:Western blot和RT-PCR测定MGC-803细胞和GES-1细胞中TBX3蛋白及mRNA水平。将MGC-803细胞分为空白对照组(BC组)、阴性对照组(NC组)、siR-TBX3组和siR-TBX3+XAV939组。CCK8测定MGC-803细胞增殖,Transwell实验检测MGC-803细胞侵袭和迁移能力。Western blot测定MGC-803细胞神经-钙黏蛋白(N-cadherin)、上皮-钙黏蛋白(E-cadherin)、Snail、Wnt3α、β-catenin、C-myc蛋白水平。结果:胃癌细胞MGC-803中TBX3蛋白和mRNA水平高于正常胃黏膜细胞GES-1(P<0.05)。与BC组和NC组相比,siR-TBX3组和siR-TBX3+XAV939组MGC-803细胞中TBX3蛋白和mRNA水平降低(P<0.05),增殖能力降低(P<0.05),细胞侵袭细胞数和迁移细胞数减少(P<0.05),N-cadherin、Snail、Wnt3α、β-catenin、C-myc蛋白水平降低(P<0.05),E-cadherin蛋白水平升高(P<0.05);与siR-TBX3组相比,siR-TBX3+XAV939组MGC-803细胞增殖能力降低(P<0.05),细胞侵袭细胞数和迁移细胞数减少(P<0.05),N-cadherin、Snail、Wnt3α、β-catenin、C-myc蛋白水平降低(P<0.05),E-cadherin蛋白水平升高(P<0.05)。BC组和NC组MGC-803细胞中各指标差异无统计学意义(P>0.05)。结论:沉默TBX3可通过抑制Wnt/β-catenin信号通路抑制胃癌细胞增殖、侵袭和迁移。 Objective:To investigate effects of TBX3 on proliferation,invasion,migration and Wnt/β-catenin signaling pathway in gastric cancer cells in order to elucidate possible mechanism of TBX3 on gastric cancer.Methods:Levels of TBX3 protein and mRNA in MGC-803 cells and GES-1 cells were determined by Western blot and RT-PCR.MGC-803 cells were divided into blank control group(BC group),negative control group(NC group),siR-TBX3 group and siR-TBX3+XAV939 group.CCK8 was used to detect MGC-803 cell proliferation.Transwell was used to measure invasion and migration of MGC-803 cells.Western blot was used to detect levels of N-cadherin,E-cadherin,Snail,Wnt3α,β-catenin and C-myc proteins in MGC-803 cells.Results:Levels of TBX3 protein and mRNA in gastric cancer cell MGC-803 were higher than those in normal gastric mucosal cells GES-1(P<0.05).Compared with BC group and NC group,TBX3 protein and mRNA levels were decreased(P<0.05),proliferation ability were decreased(P<0.05),numbers of invasion while migration cells were decreased(P<0.05),levels of N-cadherin,Snail,Wnt3α,β-catenin and C-myc protein were decreased(P<0.05) while level of E-cadherin protein was increased(P<0.05) in siR-TBX3 group and siR-TBX3+XAV939 group.Compared with siR-TBX3 group,proliferation ability were decreased(P<0.05),numbers of invasion and migration cells were decreased(P<0.05),levels of N-cadherin,Snail,Wnt3α,β-catenin and C-myc protein were decreased(P<0.05) and level of E-cadherin protein was increased(P<0.05) in siR-TBX3+XAV939 group.There were no significant difference in the indexes of MGC-803 cells between BC group and NC group(P>0.05).Conclusion:Silencing TBX3 can inhibit proliferation,invasion and migration of gastric cancer cells by inhibiting Wnt/β-catenin signaling pathway.
