摘要
目的探讨白细胞介素-6(IL-6)与磷酸化信号转导和转录激活因子3(p-STAT3)在子宫内膜癌中表达及相关性研究。方法取61例子宫内膜癌患者的子宫内膜癌组织,以及20例子宫良性病变患者的正常子宫内膜组织,免疫组化法检测不同组织中IL-6和两种p-STAT3[磷酸化STAT3-酪氨酸705(pSTAT3-Tyr705)和磷酸化STAT3-丝氨酸727(pSTAT3-Ser727)]的免疫组化评分,Spearman等级相关分析子宫内膜癌患者子宫内膜癌组织中IL-6与pSTAT3-Tyr705、pSTAT3-Ser727表达的相关性。结果正常子宫内膜组织中IL-6、pSTAT3-Tyr705、pSTAT3-Ser727蛋白的免疫组化评分均明显低于子宫内膜癌组织,差异均有统计学意义(P﹤0.01)。Spearman相关分析结果显示,IL-6的表达与pSTAT3-Tyr705、pSTAT3-Ser727的表达均相关(r=0.512、0.593,P﹤0.01)。结论子宫内膜癌中,IL-6可能通过激活STAT3信号通路促进其发生发展,提示IL-6和STAT3是子宫内膜癌的重要分子治疗靶点。
Objective To investigate the expression and correlation of interleukin-6(IL-6)with phosphorylated signal transducer and activator of transcription 3(p-STAT3)in endometrial cancer.Method 61 cases of endometrial cancer tissues and 20 cases of normal endometrial tissues were extracted,and the immunohistochemical scores of IL-6 and two kinds of p-STAT3[STAT3 phosphorylation-tyrosine 705(pSTAT3-Tyr705)and STAT3 phosphorylation-serine 727(pSTAT3-Ser727)]in different tissues were detected by immunohistochemistry.Spearman rank correlation analysis was used to analyze the expression and correlation of IL-6 with pSTAT3-Tyr705 and pSTAT3-Ser727 in endometrial cancer tissues of endometrial cancer patients.Result The immunohistochemical scores of IL-6,pSTAT3-Tyr705 and pSTAT3-Ser727 in normal endometrial tissues were significantly lower than those in endometrial cancer tissues(P<0.01).Spearman correlation analysis showed that the expression of IL-6 was correlated with the expression of pSTAT3-Tyr705 and pSTAT3-Ser727(r=0.512,0.593,P<0.01).Conclusion IL-6 may promote the occurrence and development of endometrial cancerby activating STAT3 signaling pathway,suggesting that IL-6 and STAT3 are important molecular therapeutic targets for endometrial cancer.
