摘要
目的建立高效液相色谱法测定四臣止咳颗粒中胡麻属苷、山栀苷甲酯、岩白菜素、绿原酸、甘草苷及甘草酸铵的含量。方法对四臣止咳颗粒75%甲醇提取物进行分析,采用XSelect?HSS T3柱(4.6 mm×150 mm,5μm)色谱柱;流动相为0.1%磷酸水(A)-乙腈(B),梯度洗脱;柱温20℃;流速0.8 mL·min-1;检测波长252 nm。结果胡麻属苷、山栀苷甲酯、岩白菜素、绿原酸、甘草苷及甘草酸铵的质量浓度分别在5.01~80.02μg·mL^-1(r=0.9998)、5.69~91.01μg·mL^-1(r=0.9998)、56.48~903.66μg·mL^-1(r=0.9999)、29.55~472.85μg·mL^-1(r=0.9995)、16.02~256.27μg·mL^-1(r=0.9991)、30.94~495.04μg·mL^-1(r=0.9995)与峰面积呈良好的线性关系,平均加样回收率分别为92.0%、92.6%、97.1%、99.5%、97.7%、100.2%,RSD值分别为1.3%、1.3%、1.1%、2.8%、2.9%、2.7%。结论该方法准确、重复性好,可用于四臣止咳颗粒多成分的质量控制研究。
Objective To determine sesamoside,shanzhiside methyl ester,bergenin,chlorogenic acid,liquiritin,and monoammonium glycyrrhizinate in Sichen Zhike granules by HPLC.Methods The 75%methanol extract of Sichen Zhike granules was analyzed by XSelect~?HSS T3 column(4.6 mm×150 mm,5μm).Phosphoric acid water(0.1%)(A)and acetonitrile(B)were used as the mobile phase for gradient elution with the flow rate of 0.8 mL·min-~1.The column temperature was 20℃,the injection volume was 5μL,and the detection wavelength was 252 nm.Results The concentrations of sesamoside,shanzhiside methyl ester,bergenin,chlorogenic acid,liquiritin,and monoammonium glycyrrhizinate showed good linearity at 5.01~80.02μg·mL^-1(r=0.9998),5.69~91.01μg·mL^-1(r=0.9998),56.48~903.66μg·mL^-1(r=0.9999),29.55~472.85μg·mL^-1(r=0.9995),16.02~256.27μg·mL^-1(r=0.9991),and 30.94~495.04μg·mL^-1(r=0.9995).The average recoveries of the 6 components were 92.0%,92.6%,97.1%,99.5%,97.7%and 100.2%,with the RSD of 1.3%,1.3%,1.1%,2.8%,2.9%and 2.7%,respectively.Conclusion This method is accurate and reproducible,which can be used for the multi-component quality control of Sichen Zhike granules.
