高效液相色谱法测定五维甘草那敏胶囊中泛酸钙、生物素的含量

HPLC determination of calcium pantothenate and biotin in pentavitamin licorice and chlorphenamine maleate capsules

目的建立高效液相色谱法同时测定五维甘草那敏胶囊中泛酸钙、生物素的含量。方法采用Agilent Eclipse Plus C18色谱柱(4.6 mm×250 mm,5μm);流动相为V甲醇∶V0.05%三氟乙酸溶液=25∶75;进样量为10μL;柱温为35℃;流速为1.0 mL·min-1;检测波长为200 nm。结果泛酸钙的线性范围为4.015~24.09μg(r=0.9996),高、中、低浓度的平均回收率分别为98.8%、98.9%、99.3%,RSD分别为0.75%,0.61%和0.80%;生物素的线性范围为0.0505~0.303μg(r=0.9994),高、中、低浓度的平均回收率分别为98.9%、98.3%、98.9%,RSD分别为1.4%、0.38%、1.0%。结论该方法操作简单,结果准确,重复性好,可有效地控制五维甘草那敏胶囊的质量。

Objective To simultaneously determine the content of calcium pantothenate and biotin in pentavitamin licorice and chlorphenamine maleate capsules by HPLC.Methods Agilent Eclipse Plus C18 column(4.6 mm×250 mm,5μm)was used.The mobile phase was methanol-0.05%trifluoroacetic acid solution(25∶75),the injection volume was 10μL,the column temperature was 35℃,the flow rate was 1.0 mL·min^-1,and the detection wavelength was 200 nm.Results The linearity of calcium pantothenate was 4.015~24.09μg(r=0.9996),the average recovery of high,middle,and low dose was 98.8%,98.9%,and 99.3%,with RSDs of 0.75%,0.61%and 0.80%.The linearity of biotin was 0.0505~0.303μg(r=0.9994),the average recovery of high,middle,low dose was 98.9%,98.3%,and 98.9%,with RSDs of 1.4%,0.38%,and 1.0%.Conclusion The method is simple,accurate,and repeatable,which can be used to control the quality of pentavitamin licorice and chlorphenamine maleate capsules.
