摘要 目的研究p38MAPK在正常豚鼠耳蜗中的表达。方法将10只听力正常的豚鼠取材,运用实时荧光定量PCR(qRT-PCR)和免疫组织化学技术检测p38MAPK在豚鼠耳蜗各部位中的表达差异。结果 qRT-PCR:p38MAPK mRNA在正常豚鼠耳蜗中的表达量为1.0733±0.2608;免疫组化:P38MAPK在豚鼠耳蜗中广泛表达于血管纹、螺旋韧带、前庭膜、Corti器、螺旋缘、螺旋神经节,其中螺旋神经节相对于其他部位表达较弱,差异具有统计学意义(P<0.001),余部位表达无明显差异(P>0.05)。结论 p38MAPK在豚鼠耳蜗中有表达且表达部位与AQP2及ENaC在耳蜗表达部位大致重合,可能参与膜迷路积水的形成。 Objective To study expression of p38MAPK in the cochlea of normal guinea pigs.Methods Ten normal guinea pigs were used to detect p38MAPK expression in different parts of guinea pig cochlea by real-time quantitative PCR(qRT-PCR)and immunohistochemistry.Results qRT-PCR:The mRNA relative expression of p38MAPK in the guinea pig cochlea was 1.0733±0.2608.Immunohistochemistry:P38MAPK was widely expressed in the guinea pig cochlea,including stria vascularis,spiral ligament,Reissner’s membrane,organ of Corti,spiral margin,spiral limbus and spiral ganglion,although weakly expressed in the spiral ganglion relative to other parts(P<0.001).There was no significant difference among the remaining parts(P>0.05).Conclusion p38MAPK expression approximately overlaps that of AQP2 and ENaC in the cochlea,and may be involved in the formation of endolymphatic hydrops.
