鸡蛋清水解物对亚硫酸钠诱发的疲劳斑马鱼的改善作用 被引量:1

摘要 研究了鸡蛋清水解物(HEpep)对斑马鱼的疲劳改善作用。将斑马鱼随机分为6组,分别为空白对照组(养鱼用水处理)、疲劳模型对照组、中华跌打丸组和3个HEpep浓度组。28℃培养箱中培养一段时间后,除空白对照组外,其余组均同时水溶给予亚硫酸钠以诱发斑马鱼疲劳模型,然后水溶给予中华跌打丸(1 mg/mL)处理阳性对照组,分别以水溶给予111,333,1000μg/mL质量浓度的HEpep处理HEpep剂量组。养鱼一段时间后,利用行为分析仪测定斑马鱼的运动总距离,并用凝胶渗透色谱测定了HEpep的分子量和氨基酸含量。结果表明,HEpep分子量668,303的肽段占比最大,分别占25%,48%;必需氨基酸占氨基酸总量接近36%,必需氨基酸与非必需氨基酸比值为0.56;支链氨基酸(亮氨酸+异亮氨酸+缬氨酸)含量丰富,高达15.93 mg/g。与疲劳模型对照组比较,中华跌打丸组运动改善作用为115.2%;在111,333,1000μg/mL质量浓度的条件下,其运动改善作用分别为72.5%,16.9%,1.9%,乳酸含量分别降低63.2%,18.8%,5.1%。结果表明,HEpep对亚硫酸钠诱发的疲劳斑马鱼运动能力有明显的改善作用,可以显著性降低斑马鱼体内乳酸含量。HEpep作为疲劳改善剂使用时,推荐适宜质量浓度范围为0~111μg/mL。 Effect of egg white hydroysate(HEpep)on fatigue zebrafish induced by sodium sulfite was investigated.Zebrafish were randomly divided into six groups:blank group,fatigue model group,Zhonghua Dieda pill group and three concentrations of HEpep group.After incubation at 28℃,except for the blank group,all the other groups were treated by sodium sulfite to induce zebrafish fatigue model.Then Zhonghua Dieda pill at a dose of 1 mg/mL,and HEpep at doses of 111,333 and 1000μg/mL was used to treat the fatigue zebrafish respectively.After incubation,the total movement distance of zebrafish was measured by behavior analyzer.The molecular weight and amino acid content of HEpep were also measured.The results showed that molecular weight of 668 and 303 in HEpep accounted for the largest,25%and 48%respectively;The proportion of essential amino acids to total amino acids was close to 36%,the proportion of essential amino acids to non-essential amino acids was 0.56,branched chain amino acids(Leu+Ile+Val)were rich in HEpep,up to 15.93 mg/g.The total movement distance of zebrafish in Zhonghua Dieda pill group was 115.2%of improvement compared with the model group;In the group treated by HEpep at doses of 111,333 and 1000μg/mL,the improvement rate of movement distance was 72.5%,16.9%and 1.9%respectively,the decrease rate of lactic acids content was 63.2%,18.8%and 5.1%respectively.The results indicated that HEpep could significantly improve the exercise ability and decrease lactic acid content of fatigue zebrafish induced by sodium sulfite.The optimum concentration of HEpep was 0~111μg/mL when it's used as a fatigue improver.
