摘要
为深入研究甜瓜抗白粉病的分子机制,本文在蛋白质水平上探讨甜瓜应对白粉病胁迫的可能机理,寻找特定的白粉病胁迫应激反应蛋白,为分离、鉴定和克隆抗白粉病的基因奠定基础.本研究采用蛋白质双向电泳技术(twodimensional gel electrophoresis,2-DE),对甜瓜白粉病的感病品种"JS"和抗病品种"MR-1"叶片中差异超过1.5倍的36个蛋白质点进行MALDI-TOF-TOF/MS分析和数据库检索.鉴定出31个差异蛋白质,将鉴定出的可能与甜瓜白粉病相关的差异蛋白进行了初步分析.结果表明:白粉病诱导处理对两种甜瓜的叶片蛋白质表达有明显的影响,两种甜瓜的电泳图谱间存在较为明显的差异.相比而言",MR-1"中表达量下调的蛋白质点数要明显少于"JS",而表达量上调和特异性表达的蛋白质点则明显多于"JS",这种表达差异的存在可能是"MR-1"具有较强抗性的一种重要生理响应.
In order to further study the molecular mechanism of resistance of muskmelon under the stress of powdery mildew, this paper intends to investigate the possible coping mechanism of muskmelon under powdery mildew stress in order to reveal specific protein response to powdery mildew. The results of this study will lay the foundation for isolation, identification and cloning of powdery mildew resistance gene(PMRE). Two varieties of the muskmelon were picked up: one is the disease-infected variety named "JS" and the other, disease-resistant variety called "MR-1" . The method of two-dimensional electrophoresis(2-DE) was carried out to explore the effects of proteins in the melon seedling leaves of these two varieties under powdery mildew stress,the36 protein spots where the difference between these two varieties was significant(more than 1.5 times) were analyzed with MALDI-TOFTOF/MS. After a database searching, 31 different proteins were identified, this study preliminarily analyzed the differentially identified proteins possibly related to powdery mildew of melon. The results show that the treatment of powdery mildew stress brought obvious influence on the protein expression of two kinds of melon leaves. There are significant differences between the 2-DE images of two kinds of melons. In comparison, protein spots downregulated in "MR-1" are obviously less than that in "JS" , while protein spots up-regulated and expressed specifically in "MR-1" are significantly more than that in "JS" . The obvious differences of protein expression between the two kinds of melons might reveal that "MR-1" has an important physiological response with stronger resistance than "JS" under the stress of powdery mildew.
