摘要
目的比较不同解剖部位来源脂肪干细胞在促进移植脂肪存活方面的作用差异。方法体外分离和培养来自人腹部和大腿的脂肪干细胞。取第3代脂肪干细胞,将脂肪干细胞与脂肪组织混合移植于7只裸鼠背部,每只裸鼠背部随机注入3组移植物:腹部来源脂肪干细胞辅助脂肪移植组(A组)、大腿来源脂肪干细胞辅助脂肪移植组(B组)及阴性对照组(C组)。术后3个月观察移植物情况,进行移植物体积和湿重测量。结果术后第3个月,A组、B组和C组移植物体积分别为(0.065±0.008)cm^3、(0.032±0.006)cm^3和(0.011±0.004)cm^3。实验组和阴性对照组移植物体积差异具有统计学意义(均P<0.01),A组与B组两组间比较,移植物体积差异亦具有统计学意义(P<0.05)。A组和B组脂肪移植物湿重分别为(0.201±0.020)g和(0.146±0.070)g,差异亦具有统计学意义(P<0.05)。结论与来自大腿的脂肪干细胞比较,来自腹部的脂肪干细胞可能更有利于促进腹部游离颗粒脂肪的存活。
Objective To compare the effect of adipose-derived stem cells(ADSCs)from different regions on the survival rate of free fat transplantation.Methods ADSCs from abdomen and thigh were isolated and cultured in vitro.The passage 3 ADSCs were respectively mixed with free fat graft from aspirates,then the mixtures were injected subcutaneously at the three random points on the back of seven nude mice:ADSCs from abdomen-assisted lipotransfer(A group),ADSCs from thigh-assisted lipotransfer(B group)and negative control group(C group).Transplanted fat tissue was harvested after 3 months.The volume and wet weight of transplanted fat tissue was determined.Results The volume of transplanted fat tissue was(0.065±0.008)cm^3,(0.032±0.006)cm^3 and(0.011±0.004)cm^3 in A group,B group and negative control group,respectively,with a significant difference between experimental group and negative control group(P<0.01),and a significant difference between A group and B group(P<0.05).The wet weight of transplanted fat tissue was(0.201±0.020)g and(0.146±0.070)g,with a significant difference between them(P<0.05).Conclusion Compared to ADSCs from thigh,ADSCs from abdomen have more advantage in increasing the survival rate of free fat transplantation in nude mice,indicating their potential clinical value in free fat transplantation.
