摘要
目的探讨阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者外周血Th17细胞相关细胞因子的表达水平及临床意义。方法选取2018年1月至2018年12月在武汉市第四医院耳鼻咽喉科行呼吸睡眠监测的患者55例,根据睡眠呼吸暂停指数(AHI)高低分为3组:正常对照组15例(AHI≤5次/h);轻度组20例(5<AHI<15次/h),中重度组20例(AHI≥15次/h)。采用酶联免疫吸附法(ELISA)检测受试者外周血IL-17、IL-6、IL-23水平,免疫印迹法(Western blot)检测外周血淋巴细胞中Th17细胞核转录因子RORγt蛋白的表达水平,并与代表OSAHS严重程度的参数AHI进行相关性分析。结果OSAHS中重度组患者血清IL-17、IL-6、IL-23水平显著高于轻度组及正常对照组,差异有统计学意义(均P<0.01);OSAHS中重度组患者外周血淋巴细胞RORγt蛋白表达量显著高于轻度组及正常对照组,差异有统计学意义(均P<0.01);IL-17、IL-6、IL-23水平及RORγt的蛋白表达量与OSAHS患者AHI均呈正相关(r值分别为0.95、0.87、0.92、0.91,均P<0.01)。结论OSAHS患者外周血中Th17细胞相关细胞因子表达增加,Th17细胞相关细胞因子与OSAHS严重程度相关,提示其可能参与了OSAHS的发生发展。
Objective To investigate the expression and significance of Th17 cells related cytokines in peripheral blood of patients with obstructive sleep apnea-hypopnea syndrome.Methods Totally,55 patients who underwent respiratory and sleep monitoring in our department from January 2018 to December 2018 were selected and divided into three groups according to the AHI index:15 patients in the normal control group(AHI<5 times/h);20 cases in the mild group(5<AHI<15 times/h)and 20 cases in the moderate and severe group(AHI≥15 times/h).The levels of IL-17,IL-6 and IL-23 in peripheral blood were detected by enzyme-linked immunosorbent assay(ELISA).The protein expression level of RORγt was detected by Western blotting.Results The serum concentrations of IL-17,IL-6 and IL-23 in the moderate and severe groups were significantly higher than those in the mild group and the normal control group(P<0.01).The expression of RORγt protein in severe group was significantly higher than that in the mild group and normal control group(P<0.01).The levels of IL-17,IL-6,IL-23 and RORγt were positively correlated with AHI(r=0.95,0.87,0.92,0.91,P<0.01).Conclusion Th17 cells related cytokines are increased in OSAHS patients,and they are correlated with the occurrence and development of OSAHS.
