摘要
目的:研究外周血中MicroRNA-9与MicroRNA-99a联合检测对子宫内膜癌(Endometrial carcinoma,EC)的诊断价值。方法:通过实时荧光定量RT-PCR方法检测35例子宫内膜癌患者和35例正常对照外周血中MicroRNA-9与MicroRNA-99a的表达,建立ROC曲线分析二者联合检测对子宫内膜癌的诊断意义。结果:MicroRNA-9在子宫内膜癌患者外周血中与正常对照相比表达降低,正常对照的相对表达量约为子宫内膜癌患者的1.78倍(P<0.05)。MicroRNA-99a在子宫内膜癌患者外周血中与正常对照相比表达升高,子宫内膜癌患者的相对表达量约为正常对照的2.05倍(P<0.05)。外周血中MicroRNA-9与MicroRNA-99a可以区分子宫内膜癌患者和正常人群(AUC:0.79,95%CI:0.70~0.90,AUC:0.79,95%CI:0.68~0.90)。MicroRNA-9与MicroRNA-99a联合检测鉴别子宫内膜癌患者更准确(AUC:0.98,95%CI:0.93~1.00)。结论:外周血中MicroRNA-9与MicroRNA-99a联合检测对子宫内膜癌诊断方法研究具有重要意义。
Objective:To investigate whether circulating MicroRNA-9/MicroRNA-99a might act as noninvasive biomarkers for endometrial carcinoma diagnosis.Methods:Venous blood samples(6 mL)were collected from 35 patients with endometrial carcinoma and 35 healthy controls.The plasma MicroRNA-9 and MicroRNA-99a concentrations were detected by reverse transcription polymerase chain reaction.Receiver operating characteristic(ROC)curve analysis was applied to evaluate the clinical value of plasma MicroRNA-9/MicroRNA-99a for endometrial carcinoma diagnosis.Results:The plasma MicroRNA-9 expression levels in patients with endometrial carcinoma were downregulated compared with normal control,and the relative expression level in normal controls was about 1.78 times that in patients with endometrial carcinoma(P<0.05).The plasma MicroRNA-9 expression levels in patients with endometrial carcinoma were upregulated compared with normal controls,and the relative expression level in patients with endometrial carcinoma was about 2.05 times that in normal controls(P<0.05).The plasma MicroRNA-9 and MicroRNA-99a expression levels could distinguish endometrial carcinoma patients from healthy people(AUC:0.79,95%CI:0.70~0.90,AUC:0.79,95%CI:0.68~0.90).A combination of MicroRNA-9 and MicroRNA-99a was more accurate in identifying endometrial carcinoma patients(AUC:0.98,95%CI:0.93~1.00).Conclusion:Plasma MicroRNA-9/MicroRNA-99a expressions in endometrial carcinoma might act as a novel diagnostic biomarker.
