茶树PEBP基因家族结构与功能特征分析 被引量:5

摘要 磷脂酰乙醇胺结合蛋白(phosphatidyl ethanolamine binding protein, PEBP)在控制花的发育和形成过程中起着重要作用,为了解茶树PEBP基因家族在茶树开花中的作用,本研究利用生物信息学方法从茶树基因组数据库中检索出了7个PEBP基因,并对其蛋白质理化性质、基因结构、进化关系、保守功能域、启动子顺式作用元件和表达模式进行了分析。结果表明,茶树PEBP基因编码的氨基酸数相对较少,分子量较小,主要介于14.39~20.19 kD之间,大部分为碱性蛋白;对茶树PEBP基因结构进行分析,发现该基因家族中有6个成员具有4个外显子及3个内含子的结构,基因结构比较保守;系统进化分析发现,茶树PEBP基因家族可以分为FT、TFL1、MFT亚基因家族,每个亚族所含有的基因数分别为1个、3个、3个,编码的蛋白序列均含有motif 1、motif 3、motif 4等保守基序;启动子元件分析的结果显示,PEBP基因的启动子元件类型主要以光响应元件及激素响应元件为主;分析茶树PEBP基因家族在不同组织的表达情况,发现其主要存在3种类型的表达模式,其中MFT-like亚家族的基因CsMFT与CsMFT2在生殖器官及营养器官都具有非常高的表达量。本研究获取了茶树PEBP基因家族成员的序列和部分功能信息,为进一步研究PEBP基因家族参与茶树开花调控的分子机制提供了参考。 Phosphatidyl ethanolamine binding protein(PEBP) is vital to control flower development and plant architecture. Bioinformatics analysis was used to analyze tea tree genome in order to understand the PEBP gene family of Camellia sinensis in flowering. A total of 7 PEBP genes of Camellia sinensis were retrieved from genome database, then their physicochemical properties, gene structure, genetic evolutionary relationship, functional domain, promoter element and expression patterns were studied. The results showed the encoding protein had few amino acids and the molecular weight was relatively small mainly between 14.39~20.19 kD, most of which were basic proteins. Six of 7 PEBP gene families in tea tree had 4 exons and 3 introns, and the gene structure was relatively conservative. Phylogenetic analysis showed the identified PEBP genes were classified into three groups(FT, TFL1,MFT), and all contain conserved motifs(motif 1, motif 3, motif 4). The promoter element analysis showed that there were mainly light response element and hormone response element in PEBP gene family. It was found that there were mainly three expression patterns of PEBP gene family of tea in different tissues, among which the MFT-like subfamily gene CsMFT and CsMFT2 had very high expression levels in reproductive organs and vegetative organs.In this study, sequences and some functions of PEBP gene family in tea were obtained, and it would be useful to know molecular mechanism involved in the regulation of tea flowering.
