甘草附子汤对小鼠疼痛模型及寒湿证疼痛模型止痛作用比较研究 被引量:8

摘要 目的:观察甘草附子汤对寒湿证小鼠疼痛模型的止痛作用。方法:①热板疼痛实验:将符合条件的小鼠60只,采用随机数字表法分为6组,对照组、甘草附子汤组、塞来昔布组、寒湿证模型组、寒湿证甘草附子汤组、寒湿证塞来昔布组,每组10只。给药15 d,各组小鼠在末次灌胃后45 min、90 min、135 min、180 min后放入温度为(55±0.5)℃的热板仪上分别测定小鼠的痛阈值。②扭体疼痛实验:分组与给药同热板实验。各组小鼠末次灌胃30 min后,腹腔注射0.6%冰乙酸(0.1 mL/10 g),立即将小鼠单只放入笼内,观察15 min内各鼠出现扭体反应的潜伏期(注射冰乙酸后至第一次扭体反应出现的时间)和扭体次数。结果:与寒湿证模型组比较,寒湿证甘草附子汤组扭体次数减少(P<0.01),且在给药后135 min舔脚时间变长(P<0.01);与甘草附子汤组比较,寒湿证甘草附子汤组扭体次数减少、潜伏时间延长(P<0.05),且在给药后180 min舔脚时间变长(P<0.05)。结论:甘草附子汤对小鼠寒湿证疼痛模型的止痛作用较单纯小鼠疼痛模型更显著,说明方剂的治疗效果与证型关系密切,为方证相关理论提供了实验支持。 Objective:To observe the analgesic effect of Gancao Fuzi Decoction on the pain mice model with cold-damp syndrome.Methods:①Hot plate experiment:60 eligible mice were divided into 6 groups by randomnumber table method,control group,Gancao Fuzi decoction group,celecoxib group,cold-dampness syndrome model group,cold-dampness syndrome GancaoFuzi decoction group,cold-dampness syndrome celecoxib group,each group has 10.After 15 days of administration,the mice in each group were put into a hot plate instrument at a temperature of(55±0.5)℃,then the pain threshold of the mice were determined 45 min,90 min,135 min and 180 min after gavage.②Writhing experiment:Grouping and administration were the same as hot plate experiment.30 minutes after the last gavage,0.6%glacial acetic acid(0.1 mL/10 g)was injected intraperitoneally,then the mice were immediately placed in a cage,and the incubation period of writhing reaction(the time from the injection of glacial acetic acid to the first writhing reaction)and the numbers of writhing were observed within 15 minutes.Results:Compared with the cold-dampness syndrome model group,the licking time was prolonged 135 minutes after gavage and the number of writhing was reduced in the cold-dampness syndrome Gancao Fuzi decoction group(P<0.05).Compared with Gancao Fuzi decoction group,the licking time was prolonged 180 minutes after gavage,the number of writhing was reduced,and the latency was increased in the cold-dampness syndrome Gancao Fuzi decoction group(P<0.05).Conclusion:Gancao Fuzi decoction has a more significant analgesic effect on the pain model of cold-dampness syndrome in mice than the pain model,indicating that the therapeutic effect of the prescription is closely related to the syndrome type,and provides experimental support for the theory on correlation between prescriptions and syndromes.
