摘要
目的探讨乳腺癌患者唾液蛋白内转铁蛋白(TF)、间-α胰蛋白酶抑制因子重链H1(ITIH1)、α2巨球蛋白(α2M)及锌指同源框蛋白4(ZFHX4)等4个相关蛋白的水平变化规律。方法收集18例乳腺癌患者和18例健康女性作为研究对象,采集其唾液标本,采用酶联免疫吸附试验检测唾液中TF、ITIH1、α2M及ZFHX4等4个蛋白表达水平。通过STRING和KEGG数据库对上述4种蛋白进行生物信息学分析。结果乳腺癌患者唾液中TF、ZFHX4水平分别为(3.092±0.309)mg/L、(0.546±0.084)μg/L,均高于健康女性的(2.231±0.217)mg/L、(0.207±0.059)μg/L,差异均有统计学意义(t=2.281,P=0.030;t=3.310,P=0.003)。乳腺癌患者唾液中ITIH1、α2M水平分别为(0.162±0.105)μg/L、(2.448±0.218)mg/L,均低于健康女性的(1.709±0.384)μg/L、(3.568±0.258)mg/L,差异均有统计学意义(t=3.891,P=0.001;t=3.318,P=0.003)。蛋白相互作用分析发现TF和α2M具有相互作用,TF是一个核心蛋白,还与多个蛋白具有相互作用。TF存在于缺氧诱导因子-1信号通路上,该通路在细胞生长发育和生理应激以及肿瘤的发生发展过程中具有重要作用。结论唾液中TF、ITIH1、α2M及ZFHX4等4个蛋白的水平变化可能与乳腺癌的发生发展密切相关,为乳腺癌的临床诊断提供一种新的方式。
Objective To explore the changes of four tumor-related salivary protein[transferrin(TF),inter-alpha-trypsin inhibitor heavy chain H1(ITIH1),alpha-2-macroglobulin(α2M),zinc finger homeobox protein 4(ZFHX4)]in breast cancer patients.Methods Saliva samples were collected from 18 breast cancer patients and 18 healthy women,enzyme linked immunosorbent assay was used to detect the TF,ITIH1,α2M and ZFHX4 protein levels in saliva.STRING and KEGG database were used to analyze their biological information.Results The saliva levels of TF and ZFHX4 were respectively(3.092±0.309)mg/L and(0.546±0.084)μg/L in the breast cancer patients,and were respectively(2.231±0.217)mg/L and(0.207±0.059)μg/L in the healthy women(t=2.281,P=0.030;t=3.310,P=0.003).The saliva levels of ITIH1 andα2M were respectively(0.162±0.105)μg/L and(2.448±0.218)mg/L in the breast cancer patients,and were respectively(1.709±0.384)μg/L and(3.568±0.258)mg/L in the healthy women(t=3.891,P=0.001;t=3.318,P=0.003).Protein interaction analysis revealed that TF andα2M had interaction,TF was a core protein,it also interacted with multiple proteins;TF existed in the hypoxia inducible factor-1 signal pathway,which played an important role in cell growth and development,physiological stress,the occurrence and development of tumors.Conclusion of breast cancer is closely related to the changes of TF,ITIH1,α2M and ZFHX4 in saliva,which may provide a new way for the clinical diagnosis of breast cancer.
