鹅肥肝形成过程中PRIMA1基因的表达规律及其调控研究

摘要 为了揭示可提高乙酰胆碱酯酶活性、促进保护性胆碱生成的PRIMA1基因在鹅肥肝形成过程中的表达规律及其调控因素,试验采用实时荧光定量PCR方法研究了填饲不同时期对照组和填饲组鹅肝脏、腹脂和胸肌中PRIMA1基因的表达水平,以及葡萄糖、油酸、胰岛素、棕榈酸等不同因子与PPAR激动剂对鹅原代肝细胞中PRIMA1基因表达量的调控,同时还测定了填饲鹅肥肝和对照鹅正常肝脏中乙酰胆碱酯酶的活性。结果表明:与对照组比较,填饲组可极显著诱导PRIMA1基因在肝脏中的表达(P<0.01),而抑制其在腹脂和胸肌中的表达;在鹅原代肝细胞中,200 mmol/L葡萄糖和0.5 mmol/L油酸均可显著诱导PRIMA1基因的表达(P<0.05),而100 nmol/L胰岛素、0.25 mmol/L棕榈酸以及PPAR激活剂均具有抑制作用(P<0.05);此外,填饲抑制了鹅肥肝中乙酰胆碱酯酶的活性。说明PRIMA1基因参与鹅肥肝的形成,其表达量的增加可能与填饲引起的高血脂高血糖有关,且PPAR激动剂可能参与脂肪酸对PRIMA1的表达调控。 To reveal the expression pattern and regulatory factors of PRIMA1 gene which can increase acetylcholinesterase activity and promote production of protective choline in the development of goose fatty liver.The mRNA expression of PRIMA1 in liver,abdominal fat and pectoral muscle in the control group and overfed geese at different stages of overfeeding group were detected by qRT-PCR.The regulation of PRIMA1 gene expression in goose primary hepatocytes by glucose,oleic acid,insulin,palmitic acid and PPAR agonists was detected.The activity of acetylcholinesterase in fatty liver of geese and normal liver of control geese was also measured.The results showed that compared with the control group,the mRNA expression of PRIMA1 gene in liver was significantly induced in the overfeeding group(P<0.01),while its expression in abdominal fat and pectoral muscle was inhibited.In goose primary hepatocytes,the expression of PRIMA1 gene was significantly induced by 200 mmol/L glucose and 0.5 mmol/L oleic acid(P<0.05),while 100 nmol/L insulin,0.25 mmol/L palmitic acid and PPAR activators all had inhibitory effects(P<0.05).Furthermore,the activity of AchE in goose fatty liver was inhibited by overfeeding.It indicated that PRIMA1 was involved in the formation of goose fatty liver,the increase of its expression might be partially attributed to hyperglycemia and hyperlipidemia,and PPAR might participate in regulation of PRIMA1 expression by fatty acids.
