基于高通量测序技术分析笼养熊猴肠道细菌菌群多样性 被引量:1

High-throughput sequencing for analysis of intestinal bacterial flora diversity in captive Macaca assamensis

摘要 为了明确熊猴(Macaca assamensis)肠道菌群的组成,试验采用高通量测序技术对2只成年(雌雄各1只)和2只少年(雄性)笼养熊猴粪便中的细菌16S rRNA基因V3和V4区域扩增子进行分析。结果表明:本试验共获得460种OTUs聚类信息,每个样品平均(395.00±8.54)个。共注释到15个门,平均相对丰度高于1%的有4个,分别为厚壁菌门、拟杆菌门、变形菌门和螺旋体门,丰度分别为(61.66±8.47)%、(31.84±7.23)%、(3.16±1.09)%和(1.73±0.50)%。共注释到115个属,平均相对丰度高于1%的属有19个,其中占比最高的是乳酸杆菌属,其次为理研菌科RC9菌属和普雷沃菌属9,丰度分别为(14.61±3.17)%、(11.68±3.49)%和(8.69±1.03)%。笼养熊猴肠道菌群在少年雄性与成年雄性个体间差异较小,性别间差异相对较大。肠道微生物组成在门水平上与其他猕猴属基本一致,但在属水平上有较大差异。 In order to understand the gut flora composition of the Macaca assamensis,the experiments used high-throughput sequencing technology to analyze the V3 and V4 region amplicons of the bacterial 16 S rRNA gene in the feces of two adult(one female and one male)and two juvenile(male)captive individuals.The results showed that a total of 460 OTUs clustering information were obtained in this experiment,with an average of(395.00±8.54)per sample.A total of 15 Phylum were annotated,the average relative abundance of 4 Phylum were higher than 1%,they were Firmicutes,Bacteroidetes,Proteobacteria and Spirochaetes,with relative abundances of(61.66±8.47)%,(31.84±7.23)%,(3.16±1.09)%and(1.73±0.50)%respectively.A total of 115 genus were annotated,the average relative abundance of 19 genus were higher than 1%.Among them,Lactobacillus was the highest proportion,followed by Rikenellaceae 6RC9gutgroup and Prevotella9,with relative abundances of(14.61±3.17)%,(11.68±3.49)%and(8.69±1.03)%respectively.The differences in the intestinal flora between juvenile males and adult males of captive Macaca assamensiss were small,and the differences between different sexes were relatively large.The composition of intestinal microorganisms was basically the same as that of other Macaque species at the phylum level,but there were large differences in genus levels.
