护骨胶囊对大鼠骨质疏松性骨折愈合的药效作用 被引量:1

摘要 目的研究护骨胶囊对大鼠骨质疏松性骨折愈合的影响,为护骨胶囊临床使用提供参考。方法20只SPF级SD雌性大鼠,采用数字表法随机分组,4只进行假手术(Sham组),其余16只去卵巢建立骨质疏松模型,4周后随机取4只作为模型组(OVX组)处死,另外12只大鼠制作成右侧股骨骨折模型,之后随机分为对照组(模型组)及护骨胶囊给药组(给药组),每组6只。模型组灌胃给予0.9%(质量分数)氯化钠注射液,给药组灌胃护骨胶囊溶液,均为每天给药1次,持续11周。给药结束后取心脏、肝脏、肾脏及子宫并计算脏器系数,X线检查骨折愈合情况,骨形态计量学分析骨痂部位骨形成情况,甲苯胺蓝染色观察骨折部位骨细微结构变化,并进一步观察大鼠胫骨上段骨细微结构变化及骨形态计量学分析。结果与Sham组比较,去卵巢后OVX组大鼠体质量显著上升,骨量下降,子宫明显萎缩;X线检查发现,给药组骨折基本愈合,可见骨性骨痂连接骨折端,影像学评分提高;骨形态计量学分析发现给药组骨痂部位双荧光增多,骨形成明显加快;甲苯胺蓝染色发现给药组骨小梁增多且排列规律,骨痂结构更趋合理;三色法染色及静态测量发现给药组胫骨上段骨质增多。结论护骨胶囊对大鼠骨质疏松性骨折具有明显的促进骨折愈合作用。 Objective To study the effects of Hugu Capsule on the healing of osteoporotic fractures in rats,and to provide a reference for the clinical use of Hugu Capsule.Methods Totally 20 female SPF-grade SD rats were randomly selected 4 rats for sham operation(Sham group),and 16 rats for ovariectomy.Four weeks later,4 rats were randomly selected to be sacrifice as OVX group.12 rats were accepted right femur fracture,and then randomly divided into the model group and the Hugu capsule administration group(administration group).The model group were given with saline and the administration group were given with Hugu capsule,and both groups were given once a day by po.for 5 weeks.After the administration,the heart,liver,kidney and uterus were weighed for the organ coefficients evaluation.X-ray examination,toluidine blue staining and bone morphometric analysis were performed to investigate the effectiveness of Hugu on bone fracture healing.Results Compared with the Sham group,the weight of the OVX group rats significantly increased,and the uterus significantly atrophy after ovariectomy.X-ray examination showed that the fractures of the administration group had osteophyseal connection at the fracture ends.Bone morphometric analysis found that the double-fluorescence of the epiphyseal site in the administration group increased,and bone formation significantly accelerated.Toluidine blue staining revealed that the trabeculae of the administration group increased and arranged regularly,and the epiphyseal structure became more reasonable.Masson staining found bone mass in the tibia of the administration group significantly increased compared to model group.Conclusion Hugu capsule can significantly promote fracture healing in osteoporotic fractures of rats.
